Sarcophine-diol, a Chemopreventive Agent of Skin Cancer, Inhibits Cell Growth and Induces Apoptosis through Extrinsic Pathway in Human Epidermoid Carcinoma A431 Cells.

Sarcophine-diol (SD), a structural modifications of sarcophine, has shown chemopreventive effects on 7,12-dimethylbenz(a)anthracene-initiated and 12-O-tetradecanoylphorbol-13-acetate-promoted skin tumor developments in mice. Tumorigenesis is associated with uncontrolled cell growth and loss of apoptosis. In the present study, the effects of SD on cell growth and apoptosis in human epidermoid carcinoma A431 cells were determined to assess whether SD could inhibit cell growth and/or induce apoptosis, thus elucidating possible mechanism of action.

[Aerobic bacteria associated with acute appendicitis in children].

Objective: The aim of the study was to determine the microbiological profile of acute appendicitis in children.

Study Design: Prospective descriptive study including children hospitalized for acute appendicitis.

Methods: A specimen of the appendice and the peritoneal exudates (if exists) was performed intraoperatively for aerobe bacteriological examination.

Microbial metabolism of biologically active secondary metabolites from Nerium oleander L.

Ursolic acid (1) and kaempferol (3) are two major constituents of the Mediterranean plant Nerium oleander L. Microbial metabolism of (1) with Aspergillus flavus (ATCC 9170) resulted in the formation of 3-oxo-ursolic acid derivative, ursonic acid (2). On the other hand, Cunninghamella blakesleeana (ATCC 8688A) was able to convert (3) into kaempferol 3-O-beta-D-glucopyranoside (4) as well as the new natural product kaempferol 4'-sulfate (5).

Chemopreventive effects of sarcotriol on ultraviolet B-induced skin tumor development in SKH-1 hairless mice.

Sarcotriol (ST) has been shown to be chemopreventive on 7,12-dimethyl-benz(a)anthracene (DMBA) initiated and 12-O-tetradecanoylphorbol-13-acetate (TPA)-promoted skin tumor development in CD-1 mice in recent studies from our laboratory. The objective of this study was to determine the chemopreventive effects of ST on ultraviolet B (UVB)-induced skin tumor development in female SKH-1 hairless mice, an experimental model relevant to human skin cancer development, and its possible mechanisms of action. Female SKH-1 mice were divided into two groups: Control and ST treated.

Antiepileptic ceramides from the Red Sea sponge Negombata corticata.

A new antiepileptic ceramide mixture 1 was isolated from the Red Sea sponge Negombata corticata. The structures of the metabolites were determined by extensive spectroscopic analysis. The anticonvulsant activity of 1 was measured in vivo using the pentylenetetrazole-induced seizure model.

HPLC method for the quantitative determination of sarcophine, a source of cembranoids with cancer chemopreventive activity.

An accurate, reproducible and sensitive method for the quantitative determination of sarcophine in the organic extract of the Red Sea soft coral Sarcophyton species was developed and validated. Sarcophine concentration was determined by RP HPLC using ODS column. The mobile phase was made up of 70% acetonitrile in deionized water and the pH was adjusted to 3.

Inflammatory myofibroblastic tumor of paranasal sinuses.

Inflammatory myofibroblastic tumors IMTs are clinico-pathologically distinctive but biologically controversial entities; they rarely affect the head and neck region and usually follow a benign clinical course after radical excision. We reviewed the literatures of IMTs and present a rare case of IMT arising from the maxillary paranasal sinus of an 11-year-old girl. Moreover, we elaborate on the morpho-immunophenotypic characteristics of this lesion.

Microbial transformation of 5-episinuleptolide.

5-episinuleptolide (1) is an abundant norcembranolide diterpene isolated from several species of the soft coral genus Sinularia. Biocatalytic transformation studies of 1 using Streptomyces lavendulea ATCC 8664 resulted in the isolation and characterization of two new metabolites 2 and 3. Compound 2, 6alpha-hydroxy-5-episinuleptolide was produced via a stereoselective reduction of 1 and was further metabolized into compound 3 which has a 3,8-bicylized cembranoid skeleton.

Chemopreventive effects of sarcophine-diol on skin tumor development in CD-1 mice.

The objective of this study was to determine the chemopreventive effects of sarcophine-diol (SD) on 7,12-dimethylbenz(a)anthracene initiated and 12-O-tetradecanoylphorbol-13-acetate promoted skin tumor development in mice and its possible mechanisms of action. SD pretreatment significantly (P<0.05) decreased skin papilloma development during promotion phase.

Presentation and management outcome of inflammatory bowel disease in Sudan.

This is a retrospective study conducted at the National Center for Gastrointestinal and Liver disease in Khartoum, Sudan on patients diagnosed with inflammatory bowel disease (IBD) during the period between January 1990 and May 2001. The study was conducted to determine the pattern of presentation and management outcome of inflammatory bowel disease (IBD) among Sudanese patients. All patients underwent a colonoscopy/ sigmoidoscopy with histopathological diagnosis, or a barium study.

Growth-promoting effect on iron-sulfur proteins on axenic cultures of Entamoeba dispar.

A growth-promoting factor (GPF) that promotes the growth of Entamoeba dispar under axenic culture conditions was found in fractions of mitochondria (Mt), hydrogenosomes (Hg) and chloroplasts (Cp) obtained from cells of six different protozoan, mammalian and plant species. We were able to extract the GPF from the Cp-rich leaf cells of a plant (spiderwort: Commelina communis L.) in an acetone-soluble fraction as a complex of chlorophyll with low molecular weight proteins (molecular weight [MW] approximately 4,600).

Quantitative determination of latrunculins A and B in the Red Sea sponge Negombata magnifica by high performance liquid chromatography.

An accurate, reproducible and sensitive method for the quantitative determination of latrunculins A and B in the organic extract of the Red Sea sponge Negombata magnifica was developed and validated. Latrunculin A and B concentrations were determined by RP-C18-HPLC and a mobile phase consisting of acetonitrile and water (60:40, v/v). The flow rate utilized was 1 mL min(-1) and the detector was set at 235 nm.

Closed rectopexy with transanal resection for complete rectal prolapse in adults.

Many techniques have been described for repair of complete rectal prolapse in adults. The results of abdominal approaches are superior to those of perineal approaches, but they carry the risks of major abdominal surgery. Twenty-seven patients (15 females and 12 males) were included in this study, with a mean age of 46 years.

HPLC determination of certain flavonoids and terpene lactones in selected Ginkgo biloba L. phytopharmaceuticals.

Axenic cultivation of Entamoeba dispar in newly designed yeast extract-iron-gluconic acid-dihydroxyacetone-serum medium.

Yeast extract-iron-gluconic acid-dihydroxyacetone-serum medium that allows axenic cultivation of Entamoeba dispar was designed based on casein-free yeast extract-iron-serum (YI-S) medium, and the usefulness of the medium was assessed. The main differences from YI-S medium are replacement of glucose by gluconic acid, addition of dihydroxyacetone and D-galacturonic acid monohydrate, and sterilization by filtration. This medium promoted the axenic growth of 5 strains of E.

[Perineal ectopic testis: report of four paediatric cases].

Perineal ectopic testis is a rare congenital malformation in which the testis is abnormally situated between the penoscrotal raphe and the genitofemoral fold. The authors report four new cases in children aged 2 months, 6 months, 2 years and 5 years. The abnormality was associated with an inguinal hernia in one case.

The risk of other primary cancer in patients with uveal melanoma: a retrospective cohort study of a Canadian population.

Background: The incidence of second primary malignant tumours has doubled during the last 2 decades. These tumours now represent the sixth most common group of cancers. Many authors have described the presence of multiple primary cancers in patients with uveal melanoma.

Synaptophysin immunohistochemistry in the human cochlea.

Light microscopy and immunohistochemical analyses of a freshly prepared human cochlea, removed at meningioma skull base surgery, were performed with particular emphasis on synaptophysin (SY) reactivity. Synaptophysin, a 38-kDa glycoprotein, is one of the most abundant integral membrane proteins of small presynaptic vesicles and is a useful marker for sites of synaptic transmission of the efferent olivocochlear system in the cochlea. Following fixation and decalcification, cryosections of 30 microm were prepared.

Potential of pottery materials in manufacturing radioactive waste containers.

Various pottery materials were evaluated for possible use in manufacturing containers for radioactive waste. Their potential was examined from the viewpoints of the effectiveness of disposal and the changes induced in them by gamma rays. Samples of these materials were irradiated with high-energy neutrons and gamma rays in a reactor near its core.

A semi-synthetic analog of the cembranoid sarcophine.

Semisynthesis of new sarcophine derivatives with chemopreventive activity.

The natural cembranolide sarcophine (3) and its lactone ring-opened analogue (10) were oxidized using selenium dioxide under different reaction temperatures to prepare hydroxylated derivatives. Nine new compounds were obtained, six of them targeted hydroxylated derivatives. The determination of regio- and stereochemistry as well as the mechanistic considerations on the selectivity observed in these reactions are discussed on the basis of 2D NMR and molecular modeling.
