The Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)-CRISPR associated protein 9 (Cas9) system is an efficient and rapid tool for genome editing. However, its utilization in bacteria suffers challenges such as the risk of repeated recognition and cutting by Cas9. Here we established a two-step genome editing strategy using the Streptococcus pyogenes CRISPR-Cas9 system to achieve a clean mutation with only the target sites into the Escherichia coli genome.

Purpose: To evaluate the possible antagonistic effects of Lactobacillus acidophilus (La) and Bifidobacterium adolescents (Ba) on Porphyromonas gingivalis (Pg) and Fusobacteriurn nuclearum (Fn).
Methods: Cylinder plate method was used to observe the inhibition zone formation.The suspensions of periodontal pathogens were mixed with cell free solutions of probiotics respectively, to investigate the antagonistic effect on the production of probiotics.
A truncated alkaline β-mannanase from alkaliphilic Bacillus sp. N16-5 (MAN330) was expressed and secreted in Kluyveromyces cicerisporus. The recombinant engineered strain for MAN330 production was stable during 80 generations, and the maximum yield of MAN330 reached 3,795 U/ml in 15 l fermenter.
