Chimeric antigen receptor T (CAR T) cells have shown their potential in hematological malignancies and the treatment of solid tumors, especially in metastases. However, CAR T-cell therapy may carry risks of inducing severe adverse effects, which are recognized as immune-related adverse events. Here, we report two cases of severe colitis presented with refractory bloody diarrhea, which were induced by carcinoembryonic antigen (CEA)-directed CAR T therapy in the treatment of metastatic colorectal adenocarcinoma.

We designed and synthesized 27 new amide and dipeptide derivatives containing a substituted phenylalanine as negative allosteric modulators (NAMs) for the beta-2 adrenergic receptor (βAR). These analogs aimed to improve the activity of our lead compound, Cmpd-15, by introducing variations in three key regions: the meta-bromobenzyl methylbenzamide (S1), para-formamidophenylalanine (S2), and 1-cyclohexyl-1-phenylacetyl (S3) groups. The synthesis involved the Pd-catalyzed β-C(sp)-H arylation of N-acetylglycine with 1-iodo-4-substituent-benzenes as the key step.

Article Synopsis
  • The study focuses on the characteristics and outcomes of patients with mild-to-moderate ulcerative proctitis (UP) and ulcerative proctosigmoiditis (UPS), with a follow-up period of 3 years.
  • It includes a retrospective analysis of 414 patients, revealing that 70.53% had mild disease and 29.47% had moderate disease.
  • At the end of the study, a majority (76.09%) of patients showed endoscopic improvement, with better prognosis observed in younger patients and a higher risk of disease extension among those with more severe symptoms.*

Hydrogels usually are fabricated by using monomers or preexisting polymers in precursor solutions. Here, a polyelectrolyte complex biohydrogel (Bio-PEC hydrogel) made from a precursor dough, by kneading, annealing, and crosslinking the dough of two oppositely charged polysaccharides, cationic chitosan quaternary ammonium salt (HACC) and anionic sodium hyaluronate (HA), photoinitiator (α-ketoglutaric acid), crosslinker glycidyl methacrylate (GMA), and water of very small quantity is reported. Controlled kneading and annealing homogenized the dough with respect to transforming randomly distributed, individual polymer chains into tightly wound double-stranded structures, which, upon UV irradiation, covalently sparsely crosslinked into a highly entangled network and subsequently, upon fully swollen in water, results in Bio-PEC hydrogel, HACC/HA, exhibiting near-perfect elasticity, high tensile strength, and high swelling resistance.

Microorganisms inhabiting in coastal sediments significantly affect the nitrogen cycling in coastal waters and ecosystems. However, the bacterial community that related to the key active nitrogen transformation processes in intertidal transition zone are still not understood. Across a long flat intertidal zone at depths from 0 to 3 m in Daya Bay, China, the bacterial communities in sediments and their driven nitrogen cycling potential were evaluated with environmental factors and 16S rRNA sequencing.

Background: adequate bowel preparation is related to the quality of colonoscopy. Dried lemon slices can increase gastrointestinal peristalsis, which has shown potential as an adjuvant of bowel preparation. We hypothesized that the combination of dried lemon slices and polyethylene glycol (PEG) could improve the efficacy of bowel preparation and be more acceptable to participants.
