Article Synopsis
  • Mechanical stress from orthodontic forces affects the remodeling of periodontal ligaments (PDLs) and alveolar bone, particularly influencing osteoclast formation.
  • The study focused on the impact of varying compressive forces on osteoclasts derived from macrophage cell line RAW264.7, revealing that the optimal force (7 layers of glass cover slips) increased the number of multinucleated osteoclasts.
  • Gene expression analysis showed that osteoclast-associated genes were significantly upregulated under optimal compression after just 3 hours, indicating that compressive forces accelerate osteoclastogenesis in macrophages.

The effects of mechanical stress release on osteoclastogenesis may be as important as those of mechanical stress application. However, the direct effects of mechanical stress on the behavior of osteoclasts has not been thoroughly investigated and there is limited information on the results of the release from mechanical stress. In this study, the effects of mechanical stress application and its release on osteoclast differentiation were examined.
