Article Synopsis
  • Hepatitis B virus (HBV) is a widespread chronic infection in Japan, predominantly involving genotypes B and C, but there's been a gradual increase in genotype A cases over the years.
  • A nationwide survey included 4,421 chronic hepatitis B (CHB) patients to analyze the distribution of HBV genotypes and their trends across different age groups from 2000-2016.
  • Results showed that while the overall prevalence of genotype A remained stable, it rose among younger adults; meanwhile, genotypes B and C shifted to older populations, highlighting the need for future studies on the impact of Japan's universal HBV vaccination introduced in 2016.

Background: Thrombocytopenia represents an obstacle for invasive procedures in chronic liver disease (CLD) patients. We aimed to estimate the appropriate dose and evaluate the efficacy and safety of lusutrombopag for the treatment of thrombocytopenia before percutaneous liver radiofrequency ablation (RFA) for primary hepatic cancer in patients with CLD.

Methods: In this multicenter, randomized, double-blind, placebo-controlled study conducted in Japan, 61 CLD patients with platelet count < 50 × 10/µL at screening were randomized to placebo or lusutrombopag 2, 3, or 4 mg once daily for 7 days, followed by a 28-day post-treatment assessment period.

Article Synopsis
  • - The study involved a genome-wide association study (GWAS) focused on identifying genetic factors associated with HBV-related liver cancer (HCC) among Japanese individuals, including 473 HCC patients and 516 HBV carriers.
  • - Researchers found 65 significant SNPs in the HLA class I region, and confirmed three of these SNPs through further analysis, showing strong links to HCC risk in multiple East Asian populations.
  • - The study also highlighted a specific HLA allele (HLA-A 33:03) linked to disease progression to HCC, but additional genetic influences in the HLA class I region were suggested since conditioning on HLA did not eliminate the associations of the three SNPs.

The impact of body mass index (BMI) on postoperative survival in Japanese patients with pancreatic cancer is unclear. We examined the relationship between preoperative BMI and the prognosis of Japanese patients who underwent surgery for pancreatic cancer to determine whether BMI affects these patients' prognosis. Of the patients who underwent pancreatectomy between January 2004 and August 2015 at our institution, 246 were pathologically diagnosed with pancreatic tubular adenocarcinoma; the cancer was located in the pancreatic head (n=161) and in the body and tail (n=85).

Background: Hepatic arterial infusion chemotherapy plus sorafenib in phase 2 trials has shown favourable tumour control and a manageable safety profile in patients with advanced, unresectable hepatocellular carcinoma. However, no randomised phase 3 trial has tested the combination of sorafenib with continuous arterial infusion chemotherapy. We aimed to compare continuous hepatic arterial infusion chemotherapy plus sorafenib with sorafenib alone in patients with advanced, unresectable hepatocellular carcinoma.

Following the publication of this article, we realize that there were some errors in the manuscript. Details of the experiments describing the gene silencing of RUNX3 with small interfering RNA (siRNA) were erroneously included in this paper, and all references to siRNA should have been deleted from the manuscript prior to publication. In the subsection entitled 'Cell lines and cell culture' on page 2577, the left‑hand column, the text should have indicated that the human HCC cell lines Hep3B and Huh7 were obtained from the American Type Culture Collection (ATCC; Manassas, VA, USA), whereas HLF cells were obtained from the Japanese Cancer Resources Bank (Tokyo, Japan).

Background And Aim: It remains unclear whether primary biliary cholangitis (PBC) represents a risk factor for secondary osteoporosis.

Methods: A case-control study was conducted to examine bone mineral density and bone turnover markers in middle-aged postmenopausal PBC patients without liver cirrhosis. We compared the incidence of low bone mineral density between propensity-score matched subgroups of PBC patients and healthy controls and investigated the mechanisms underlying unbalanced bone turnover in terms of the associations between bone turnover markers and PBC-specific histological findings.

A previous genome-wide association study (GWAS) performed in 963 Japanese individuals (487 primary biliary cholangitis [PBC] cases and 476 healthy controls) identified TNFSF15 (rs4979462) and POU2AF1 (rs4938534) as strong susceptibility loci for PBC. In this study, we performed GWAS in additional 1,923 Japanese individuals (894 PBC cases and 1,029 healthy controls), and combined the results with the previous data. This GWAS, together with a subsequent replication study in an independent set of 7,024 Japanese individuals (512 PBC cases and 6,512 healthy controls), identified PRKCB (rs7404928) as a novel susceptibility locus for PBC (odds ratio [OR] = 1.

Article Synopsis
  • Rapid mucosal healing is crucial for patients with ulcerative colitis (UC), and this study investigates the role of Spred2, a protein that affects inflammation and cell growth in the colon.
  • Researchers found that Spred2 knockout (KO) mice showed increased cell proliferation and were less affected by colitis induced by dextran sulfate sodium (DSS), exhibiting milder symptoms and quicker recovery compared to regular mice.
  • The findings suggest Spred2 is involved in regulating inflammation and cell growth in the colon, indicating it could be a potential therapeutic target for treating UC.
