Background: The demand for nurses in advanced practice roles is expected to grow by 38% by 2032. The nationwide faculty shortage is also a concern, as it correlates to the nursing shortage. Successful retention strategies for graduate nursing students can strengthen the rising demand of specialized nursing roles.

Article Synopsis
  • Current treatments for metastatic pancreatic ductal adenocarcinoma yield very low survival rates, prompting research into new therapies like mitazalimab combined with a modified chemotherapy regimen called mFOLFIRINOX.
  • The OPTIMIZE-1 study enrolled 70 patients across 14 hospitals in Belgium, France, and Spain to evaluate the safety and effectiveness of this combination therapy, focusing on determining the optimal dose of mitazalimab as well as measuring tumor response.
  • The trial successfully determined 900 μg/kg of mitazalimab as the recommended dose for the next phase and gathered data on the initial outcomes for the patients treated within the study timeframe from September 2021 to March 2023.

CD40-targeting therapies can enhance the dendritic cell priming of tumor-specific T cells and repolarize intratumoral macrophages to alleviate the tumoral immunosuppressive environment and remodel the extracellular matrix. Mitazalimab is a potent agonistic CD40 monoclonal IgG1 antibody currently under clinical development. This study used RNA sequencing of blood samples from a subset of patients from a Phase I trial with mitazalimab (NCT02829099) to assess peripheral pharmacodynamic activity.

Background: The CTLA-4 blocking antibody ipilimumab has demonstrated substantial and durable effects in patients with melanoma. While CTLA-4 therapy, both as monotherapy and in combination with PD-1 targeting therapies, has great potential in many indications, the toxicities of the current treatment regimens may limit their use. Thus, there is a medical need for new CTLA-4 targeting therapies with improved benefit-risk profile.

People with Alzheimer's disease may be able to recall events from their past clearly yet unable to remember events from minutes or hours ago. They often hold outdated beliefs about themselves, which can trigger challenging behaviour. This article explains how the simple analogy of the "time machine" can help staff in all settings to understand belief systems in dementia.

We were the first to demonstrate that combined immunotherapy with GM-CSF producing GL261 cells and recombinant IFNgamma of preestablished GL261 gliomas could cure 90% of immunized mice. To extend these findings and to uncover the underlying mechanisms, the ensuing experiments were undertaken. We hypothesized that immunizations combining both GM-CSF and IFNgamma systemically would increase the number of immature myeloid cells, which then would mature and differentiate into dendritic cells (DCs) and macrophages, thereby augmenting tumor antigen presentation and T-cell activation.

High-grade gliomas are one of the most aggressive human tumors with <1% of patients surviving 5 years after surgery. Immunotherapy could offer a possibility to eradicate remnant tumor cells after conventional therapy. Experimental immunotherapy can induce partial cure of established intracerebral tumors in several rodent models.

Glioblastoma multiforme is the most common malignant primary brain tumor and also one of the most therapy-resistant tumors. Because of the dismal prognosis, various therapies modulating the immune system have been developed in experimental models. Previously, we have shown a 37-70% cure in a rat glioma model where rats were peripherally immunized with tumor cells producing IFNgamma.
