Data-independent acquisition (DIA) on ion mobility mass spectrometers enables deep proteome coverage and high data completeness in large-scale proteomics studies. For advanced acquisition schemes such as parallel accumulation serial fragmentation-based DIA (diaPASEF) stability of ion mobility (1/K) over time is crucial for consistent data quality. We found that minor changes in environmental air pressure systematically affect the vacuum pressure in the TIMS analyzer, causing ion mobility shifts.

Introduction: Dementia is one of the most relevant widespread diseases, with a prevalence of currently 55 million people with dementia worldwide. However, about 60-75% of people with dementia have not yet received a formal diagnosis. Asymptomatic screening of cognitive impairments using neuropsychiatric tests has been proven to efficiently enhance diagnosis rates.

Introduction: Challenging behaviour (CB) is a common issue among children with autism spectrum disorder or intellectual and developmental disability. Mental health applications are low-threshold cost-effective tools to address the lack of resources for caregivers. This pre-post study evaluated the feasibility and preliminary effectiveness of the smartphone app using algorithm-based behaviour analysis to identify causes of CB and provide individualized practical guidance to manage and prevent CB.

Background: Life situation dimensions, such as financial resources, housing, health and social relationships, have a significant influence on the scope available to older people for coping with everyday life and participation and thus for a good life in old age.

Method: As a basis for identifying current and future challenges posed by precarious living conditions in old age, current publicly available data and study results on the income situation, housing, health and care are reported as central dimensions of the living conditions of people aged 65 years and over in Germany.

Results: The study results presented provide indications as to which groups of older people live in particularly precarious living situations, which provide starting points for municipal action.

Purpose: Severe burn injuries are often accompanied by infections and associated with high morbidity and mortality. This study aimed to compare the prevalence and clinical impact of bacteremia between patients receiving intensive care with and without burns.

Methods: This single-center retrospective cohort study at the University Hospital Vienna, Austria, analyzed blood cultures from intensive care unit (ICU) patients with and without burns (2012-2022) to assess the prevalence of bacteremia, the associated pathogen distribution and the 60-day all-cause mortality.

Article Synopsis
  • This study investigates whether thrombectomy alone is as effective as combining it with intravenous thrombolysis for patients with acute ischemic stroke caused by large-vessel blockage.
  • A meta-analysis of six trials involving 2,334 patients showed that functional independence at 90 days was similar between the two treatments, with slight variations in success and risk of hemorrhage.
  • While thrombectomy alone proved to be non-inferior at a wider margin, there might still be important differences in outcomes that need further exploration.

Background: Challenging behaviour (CB) comprises various forms of aggressive and problematic behaviours frequently occurring in children with intellectual and developmental disability (IDD) or autism spectrum disorder (ASD). CB often arises from impaired communication or problem solving skills. It is often met with coercive measure due to a lack of alternative strategies on the part of the caregiver, while it also impacts on the caregivers due to the exposure to physical harm and high levels of stress.

Article Synopsis
  • COVID-19 causes a hypercoagulatory state that can lead to serious blood clot issues, and while anticoagulation helps reduce mortality, the benefits of antiplatelet therapy are uncertain.
  • A study of 578 hospitalized COVID-19 patients found no correlation between antiplatelet therapy and survival rates, with adverse outcomes more tied to an overactive coagulation system.
  • Analysis of postmortem lung biopsies and patient blood samples indicated that while clotting was problematic, platelet function appeared impaired in severe cases, suggesting that antiplatelet therapy did not improve outcomes and might even worsen platelet issues.

Objective: To develop and validate a prognostic model for in-hospital mortality after four days based on age, fever at admission and five haematological parameters routinely measured in hospitalized Covid-19 patients during the first four days after admission.

Methods: Haematological parameters measured during the first 4 days after admission were subjected to a linear mixed model to obtain patient-specific intercepts and slopes for each parameter. A prediction model was built using logistic regression with variable selection and shrinkage factor estimation supported by bootstrapping.

Thromboembolic complications are frequently observed in Coronavirus disease 2019 (COVID-19). While COVID-19 is linked to platelet dysregulation, the association between disease outcome and platelet function is less clear. We prospectively monitored platelet activation and reactivity in 97 patients during the first week of hospitalization and determined plasma markers of platelet degranulation and inflammation.

Article Synopsis
  • Anticoagulation, particularly using low-molecular-weight heparin (LMWH), was linked to better survival rates in hospitalized COVID-19 patients, with significant statistical evidence supporting this finding.
  • A study of 586 COVID-19 patients revealed that while anticoagulation did not significantly reduce markers of blood clotting increase over time, it did decrease cell death markers and shortened the duration of viral positivity.
  • The research suggests that LMWH may not directly affect blood coagulation metrics but still contributes to lower mortality and better patient outcomes in those hospitalized with COVID-19.

Faldaprevir (FDV), a substrate of CYP3A/P-glycoprotein (P-gp), is a selective inhibitor of the hepatitis C virus (HCV) NS3/4 protease. FDV is currently under clinical development for application in interferon-free treatment regimens for patients with chronic HCV infection. Understanding the drug-drug interaction potential of FDV is critical, as certain drug combinations may facilitate the more rapid achievement of steady-state-that is, the ideal drug concentration and balanced metabolic cycle of absorption and elimination that optimize drug efficacy.

Background: Studies show risks in the psychopharmacological care of older people with long-term care needs. In developing recommendations on the handling of psychotropic drugs in outpatient long-term care, the perspective of care professionals has so far received only sporadic attention.

Objective: The study aims to explore perspectives in outpatient care on the use of psychopharmaceuticals in the care of older people in need of long-term care.

Background: Studies show a variety of risks associated with psychotropic medication for older people in need of long-term care in Germany. In inpatient care, nursing has important tasks when dealing with psychotropic drugs, such as placement and administration, patient observation and communication with doctors.

Aim Of Study: The study examined experiences and assessments of the handling of psychotropic drugs in inpatient care from the perspective of managers.

For machine manufacturing companies, besides the production of high quality and reliable machines, requirements have emerged to maintain machine-related aspects through digital services. The development of such services in the field of the Industrial Internet of Things (IIoT) is dealing with solutions such as effective condition monitoring and predictive maintenance. However, appropriate data sources are needed on which digital services can be technically based.

To build, run, and maintain reliable manufacturing machines, the condition of their components has to be continuously monitored. When following a fine-grained monitoring of these machines, challenges emerge pertaining to the (1) feeding procedure of large amounts of sensor data to downstream processing components and the (2) meaningful analysis of the produced data. Regarding the latter aspect, manifold purposes are addressed by practitioners and researchers.

Background And Objectives: Schizophrenia and Alzheimer's disease are characterised by glutamatergic pathway abnormalities related to N-methyl-D-aspartate (NMDA) receptor hypofunction and cognitive impairment. Glycine is an NMDA receptor co-agonist; inhibition of glycine transporter 1 (GlyT1) should improve NMDA receptor hypofunction. This study evaluated safety and pharmacokinetic properties of BI 425809-a potent and selective GlyT1 inhibitor.
