Aberrant immune responses to viral pathogens contribute to pathogenesis, but our understanding of pathological immune responses caused by viruses within the human virome, especially at a population scale, remains limited. We analyzed whole-genome sequencing datasets of 6,321 Japanese individuals, including patients with autoimmune diseases (psoriasis vulgaris, rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), pulmonary alveolar proteinosis (PAP) or multiple sclerosis) and coronavirus disease 2019 (COVID-19), or healthy controls. We systematically quantified two constituents of the blood DNA virome, endogenous HHV-6 (eHHV-6) and anellovirus.

Objective Interventions that reduce sitting time are easier to implement than those that aim to increase physical activity in compliance with the guidelines. There is no consensus on the association between sitting time as assessed by the International Physical Activity Questionnaire (IPAQ) and physical function. We investigated the association between self-reported sitting time and physical function according to the Kihon Checklist (KCL) among community-dwelling older adults.

Neuroinflammation, a hallmark of neurodegenerative diseases, is associated with neuronal cell loss and cognitive dysfunction. Monoacylglycerol lipase (MAGL) is involved in neuroinflammation in the brain via the degradation of endocannabinoid 2-arachidonoylglycerol to arachidonic acid, a precursor of some eicosanoids; therefore, MAGL inhibitors are expected to have anti-inflammatory effects. We recently developed a reversible, selective, central nervous system penetrant, and orally available MAGL inhibitor, compound 4f.

Article Synopsis
  • - Surgery for craniosynostosis aims to increase intracranial volume and improve skull shape, with endoscope-assisted suturectomy being a minimally invasive option preferred for patients under 3 months old at the National Center for Child Health and Development in Tokyo.
  • - Despite its effectiveness, some patients may experience residual bone defects several years post-surgery, particularly in the temporal region, which can complicate future procedures like monobloc advancement.
  • - A study reviewed five patients who underwent endoscope-assisted suturectomy, finding that two had significant temporal bone defects that could hinder surgical planning for further cranial corrections, highlighting the need for careful future considerations during the initial surgery.

  • Fronto-orbital advancement (FOA) is the primary treatment for craniosynostosis and has been performed using gradual distraction techniques in Asia, but not widely adopted in the West due to potential scalp tension differences among patients.
  • A study compared skin tension measurements in 12 European and 16 Asian patients undergoing cranioplasty, revealing that European patients exhibited higher tension levels, particularly on the left and right sides.
  • The findings indicate that scalp tension varies by race, which should be taken into account when determining the best treatment approach for craniosynostosis.

This manuscript unveils the synthesis of 2-thiazolo[4,5-][1,2,3]triazole (ThTz), an unprecedented [5-5]-fused heteroaromatic system, and established a scalable synthetic procedure for producing large quantities of the ThTz ring bearing a sulfone group on the thiazole ring. The sulfone moiety proves to be a versatile reactive tag, facilitating diverse transformations such as SAr reactions, metal-catalyzed couplings, and radical-based alkylations. Furthermore, functionalization of the triazole ring highlights the potential of this newly developed heteroaromatic compound as a valuable heteroaryl building block, promoting scaffold hopping strategies in medicinal chemistry.

Article Synopsis
  • * Out of 138 adult patients who used dupilumab for over 3 months, 42% discontinued it at least once, but only 5.1% maintained remission solely with topical treatment after stopping.
  • * Maintaining remission was linked to factors like lower itch and symptom scores, longer treatment duration, and less severe skin involvement at the time of discontinuation, indicating that careful patient selection is crucial for those considering stopping dupilumab.

Background/aim: The liver effectively restores both size and function following partial hepatectomy (PHx). Angiogenesis is crucial for the repair and regeneration of liver tissue post-PHx. Calcitonin gene-related peptide (CGRP) released from sensory nerves and its receptor-receptor activity-modifying protein 1 (RAMP1) are involved in angiogenesis.

Objective: Minimally invasive radical hysterectomy has a worse prognosis than open surgery, but the reasons for the poor prognosis remain unclear. Tumor spillage occurs when the tumor is exposed to the surgical field and has been suggested to be related to a poor prognosis. This study aimed to compare the prognostic value of tumor spillage in laparoscopic radical hysterectomy and evaluate whether tumor spillage prevention improves oncological safety.

Light chain proximal tubulopathy (LCPT) is a rare type of paraprotein-related disease (PRDs) characterized by monoclonal free light chain (FLC) deposition in proximal tubular epithelial cells (PTECs). A diagnosis of LCPT requires identification of FLC deposition in PTECs; however, FLC luminescence defects in immunofluorescence staining using frozen tissue (IF-F), regarded as "masked LCPT", are occasionally encountered. We describe two cases of focal masked LCPT in monoclonal gammopathy of undetermined significance (MGUS) or smoldering multiple myeloma (SMM) diagnosed by IF in formalin-fixed, paraffin-embedded tissue sections following pronase digestion (IF-P) rather than by IF-F.

While studying transgene expression after systemic administration of lentiviral vectors, we found that splenic B cells are robustly transduced, regardless of the types of pseudotyped envelope proteins. However, the administration of two different pseudotypes resulted in transduction of two distinct B cell populations, suggesting that each pseudotype uses unique and specific receptors for its attachment and entry into splenic B cells. Single-cell RNA sequencing analysis of the transduced cells demonstrated that different pseudotypes transduce distinct B cell subpopulations characterized by specific B cell receptor (BCR) genotypes.
