The volume and composition of airway surface liquid (ASL) is regulated by liquid secretion and absorption across airway epithelia, controlling the pH, solute concentration, and biophysical properties of ASL in health and disease. Here, we developed a method integrating explanted tracheal tissue with a micro-machined device (referred to as " trachea-chip") to study the dynamic properties of ASL volume regulation. The trachea-chip allows real-time measurement of ASL transport () with intact airway anatomic structures, environmental control, high-resolution, and enhanced experimental throughput.
