Backgrounds: Neuropathic pain (NP) is a kind of chronic pain that has attracted much attention in clinical practice, characterized by high morbidity, complex mechanisms, and difficulties in clinical treatment, with which the activation of High mobility group box 1 (HMGB1) is closely related. The aim of this study was to investigate the effects of lentivirus-mediated RNA interference gene therapy targeting HMGB1 on neuropathic pain in rats with chronic dorsal root ganglion compression (CCD) and its specific mechanisms, so as to explore new pharmacological targets.

Methods: Adult male Wistar rats were surgically subjected to chronic compression of the dorsal root ganglia (CCD).

Article Synopsis
  • Neuropathic pain (NP) is linked to glial cells, but the exact role of oligodendrocytes in NP alongside microglia and astrocytes is not well understood.
  • Using a mouse model, researchers examined changes in NP and myelin after a chronic injury, analyzing glial cell interactions through single-cell sequencing.
  • Findings indicated that during NP development, oligodendrocytes were less effective in myelin production due to their differentiation being impacted by other glial cells, highlighting significant interactions that may contribute to demyelination.

A tunnel boring machine (TBM) is a type of heavy load equipment that is widely used in underground tunnel construction. The geological conditions in the tunneling process are decisive factors that directly affect the control of construction equipment. Because TBM tunneling always takes place underground, the acquisition of geological information has become a key issue in this field.
