Article Synopsis
  • - A 65-year-old male with refractory diffuse large B-cell lymphoma (R/R DLBCL) underwent CAR-T cell therapy after failing standard treatment, leading to unexpected improvements in his long-standing psoriasis.
  • - Four weeks post-CAR-T infusion, the patient experienced relief from abdominal pain and notable skin improvements, with follow-up confirming complete remission of his cancer and minimal psoriasis lesions.
  • - This case suggests that CAR-T cell therapy might offer additional benefits beyond cancer treatment, potentially paving the way for new approaches to tackle complex skin conditions like psoriasis.

Objective: To investigate the effects of down-regulation of p21 activated kinase 1 (PAK1) on the proliferation, differentiation, and apoptosis of myeloproliferative neoplasm (MPN) cells (6133/MPL) with thrombopoietin receptor MPL mutation at codon 515 () and survival of 6133/MPL mice.

Methods: Interference with the protein level of in 6133/MPL cells was assessed using lentivirus-mediated shRNA transfection technology. CCK-8 assay was used to detect the effect of down-regulation of on the proliferation viability of 6133/MPL cells, and colony-forming ability was measured by cell counting.

Article Synopsis
  • The study aimed to evaluate how endothelial activation and stress index (EASIX) affect the prognosis of patients with angioimmunoblastic T-cell lymphoma (AITL) and peripheral T-cell lymphoma, not otherwise specified (PTCL-NOS).
  • Researchers analyzed clinical data from 59 recently diagnosed patients to determine an optimal EASIX cut-off value and assess survival outcomes.
  • Findings revealed that a higher EASIX (≥0.95) correlates with more severe clinical features and significantly shorter overall survival and progression-free survival, indicating it as an independent risk factor for worsened prognosis.

Risk-stratification of acute myeloid leukemia (AML) based on (cyto)genetic aberrations, including hotspot mutations, deletions and point mutations have evolved substantially in recent years. With the development of next-generation sequence technology, more and more novel mutations in the AML were identified. Thus, to unravel roles and mechanism of novel mutations would improve prognostic and predictive abilities.

Polo-like kinase 4 (PLK4), a key regulator of centriole biogenesis, is frequently overexpressed in cancer cells. However, roles and the mechanism of PLK4 in the leukemiagenesis of acute myeloid leukemia (AML) remain unclear. In this study, the PLK4 inhibitor Centrinone and the shRNA knockdown were used to investigate roles and the mechanism of PLK4 in the leukemiagenesis of AML.

Purpose: FLT3 mutations occurred in approximately one third of patients with acute myeloid leukemia (AML). FLT3-ITD mutations caused the constitutive activation of the RAS/MAPK signaling pathway. Ribosomal S6 Kinases (RSKs) were serine/threonine kinases that function downstream of the Ras/Raf/MEK/ERK signaling pathway.

Background: Whether combined CsA with androgen therapy was superior to androgen therapy alone in NSAA remains controversial. This study aimed to assess the efficacy and safety of combined therapy versus androgen therapy for NSAA patients using a meta-analytic approach.

Methods: An electronic database of PubMed, EmBase, Cochrane library, CNKI, VIP, and Wanfang was systematically searched for randomized controlled trials (RCTs) from their inception to February 2020.

The objective of this study was to assess the effects of dietary supplementation of keratinase on the production of broilers fed a diet containing feather meal. A total of 162 1-d-old Cobb 500 male broiler (n = 9 cages/diet with 6 chicks/cage) were randomly allocated to 3 dietary treatments. The broilers were fed a corn-soybean-feather meal based diet (BD), or BD supplemented with keratinase at 100,000 or 200,000 U/kg for 6 weeks.

Our previous studies have implicated Caspase-1 signaling in driving the proinflammatory state of acute graft versus host disease (aGVHD). Therefore, we aimed to elucidate the mechanism of Caspase-1 in in murine models of aGVHD through specific inhibition of its activity with the decoy peptide Ac-YVAD-CMK. We transplanted bone marrow from donor C57BL/6 (H-2b) mice into recipient BALB/c (H-2Kd) mice and randomized the recipients into the following treatment cohorts: (1) allogeneic hematopoietic stem cell transplantation and splenic cell infusion control (PBS group); (2) low dose Ac-YVAD-CMK (AC low group); (3) and high dose Ac-YVAD-CMK (AC high group).

Article Synopsis
  • The study aimed to explore the expression and clinical relevance of VCAN and related molecules in patients with multiple myeloma (MM).
  • After analyzing bone marrow samples from 25 MM patients, results showed that the expression of VCAN, FAK, and FN significantly decreased following treatment, while MK and HAS levels remained unchanged.
  • Furthermore, higher VCAN levels correlated with poor patient outcomes, suggesting it could serve as a potential biomarker for monitoring disease progression in multiple myeloma.
