Immune deficits after CD19 chimeric antigen receptor (CAR) T-cell therapy can be long-lasting, predisposing patients to infections and non-relapse mortality. In B-cell non-Hodgkin lymphoma (B-NHL), the prognostic impact of immune reconstitution (IR) remains ill-defined, and detailed cross-product comparisons have not been performed to date. In this retrospective observational study, we longitudinally characterized lymphocyte subsets and immunoglobulin levels in 105 B-NHL patients to assess patterns of immune recovery arising after CD19 CAR-T.

Background: Cranial measurements are crucial for evaluating preterm general development because they are a surrogate tool for evaluating brain growth. Usually, they are based on tape-measured head circumference; however, a three-dimensional (3D) approach expands the diagnostic spectrum to the evaluation of cranial volume and shape.

Aims: Very preterm (vPT) infants face multiple risks and obstacles in their early development.

Article Synopsis
  • In kidney transplant recipients, BK polyomavirus-associated nephropathy (BKPyVAN) is a significant risk for transplant loss, and reducing immunosuppression to manage BKPyV-DNAemia can also heighten the risk of graft rejection.
  • The CERTAIN study, involving 195 pediatric kidney transplant recipients, found that BKPyV-DNAemia significantly increased the risk of T cell-mediated rejection, development of donor-specific antibodies, and overall decline in graft function.
  • The findings suggest that careful monitoring and regular screening for donor-specific antibodies are essential when adjusting immunosuppressive therapy in patients with BKPyV-DNAemia to mitigate rejection risks.

Background: Health-related quality of life (HRQL) has become an important outcome parameter in cardiology. The MOS 36-ltem Short-Form Health Survey (SF-36) and the PROMIS-29 are two widely used generic measures providing composite HRQL scores. The domains of the SF-36, a well-established instrument utilized for several decades, can be aggregated to physical (PCS) and mental (MCS) component summary scores.

The basal structure of the bacterial flagellum includes a membrane embedded MS-ring (formed by multiple copies of FliF) and a cytoplasmic C-ring (composed of proteins FliG, FliM and FliN). The SRP-type GTPase FlhF is required for directing the initial flagellar protein FliF to the cell pole, but the mechanisms are unclear. Here, we show that FlhF anchors developing flagellar structures to the polar landmark protein HubP/FimV, thereby restricting their formation to the cell pole.

  • The Mycobacterium avium complex (MAC) includes 12 species but focuses on M. avium, M. intracellulare subsp. intracellulare, and M. intracellulare subsp. chimaera, which are significant for clinical relevance in Central Europe.
  • Whole genome sequencing was performed on 610 MAC isolates from different countries, allowing for phylogenetic analysis and identification of resistance and virulence genes.
  • Results showed clustering of isolates with minimal SNP differences, but no clear correlation between genetic data and clinical outcomes, with specific species being less common in cases of extra-pulmonary disease.

The number of adults with congenital heart defects (CHD) is steadily rising and amounts to approximately 360,000 in Germany. CHD is often associated with pulmonary hypertension (PH), which may develop early in untreated CHD. Despite timely treatment of CHD, PH not infrequently persists or recurs in older age and is associated with significant morbidity and mortality.

Article Synopsis
  • Researchers wanted to know if hospital data on sepsis (a serious infection) was correct enough to help make decisions about healthcare.
  • They checked records from 10,334 patients in 10 hospitals in Germany to see if the doctors correctly mentioned sepsis in their notes.
  • They found that sometimes sepsis was missed or not written down, which means earlier reports of how many people had sepsis were too low, and hospitals varied a lot in how well they identified it.

Several studies suggested that transcription factor (TF) binding to DNA may be impaired or enhanced by DNA methylation. We present MeDeMo, a toolbox for TF motif analysis that combines information about DNA methylation with models capturing intra-motif dependencies. In a large-scale study using ChIP-seq data for 335 TFs, we identify novel TFs that show a binding behaviour associated with DNA methylation.

The subjective impact of the consequences of pediatric traumatic brain injury (pTBI) on different life dimensions should be assessed multidimensionally and as sensitively as possible using a disease-specific health-related quality of life (HRQoL) instrument. The development and psychometrics of the first such self-report questionnaire for children and adolescents after TBI are reported here. Focus group interviews with children, adolescents, and their parents, cognitive debriefing, item pool generation and reduction using Delphi expert panels were performed.

In recent years, the selective inhibition of FKBP51 has emerged as a possible treatment for chronic pain, obesity-induced diabetes, or depression. All currently known advanced FKBP51-selective inhibitors, including the widely used SAFit2, contain a cyclohexyl residue as a key motif for enabling selectivity over the closest homologue and anti-target FKBP52. During a structure-based SAR exploration, we surprisingly discovered thiophenes as highly efficient cyclohexyl replacement moieties that retain the strong selectivity of SAFit-type inhibitors for FKBP51 over FKBP52.
