Experiments are described which probe the relationship between three sequence elements which make up the eukaryotic RNA polymerase II promoter. A cloned eukaryotic gene, from which the TATA-box and 400 base pairs of 5'-flanking sequence has been deleted, is still transcriptionally active in vivo (following its transfection into cultured mammalian cells) and in vitro. Deletion has appropriately positioned a cluster of five TATA box-like sequences upstream from multiple potential cap sites.

We have studied how small changes in the distance between the TATA-box and cap site affect transcription of a eukaryotic gene in vitro and in vivo. The trout protamine gene TPG-3 [Gregory et al. (1982) Nucl.

This paper presents three cases of serologically documented viral retinitis, and the great value of fluorescein angiography in outlining the structural abnormalities and the site of the lesions. Of our series in the case of influenza retinitis, the fluoroangiographic findings showed dye leakage from retinal vessels in the posterior pole. The dye leakage did not appear completely in the sites of macular edema.

Some restriction endonucleases generate a single-stranded nick at their recognition sequences in the presence of ethidium bromide (EtBr). This nick can then be extended to a single-stranded gap in which mutations can be introduced by a variety of techniques. To date, the templates used in these studies have largely contained a single recognition site for a given enzyme.

Sixty-five patients with carcinoma in situ of the vulva were reviewed. Attention was focused upon the epidemiology, duration of symptoms, and location of lesions. The vulva and perineum were divided diagrammatically into 16 geographic compartments; lesion location and percentage of occurrence in each area were calculated.

The mechanical properties of the lung (elasticity, laminar and turbulent resistance) were measured in 14 patients one day before and after abdominal surgery. A special measuring instrument was made for this purpose. The comparison of the conditions before and after operation showed that elasticity and turbulent resistance do not change significantly, but the value of laminar resistance showed an increase caused by the qualitative change of the lung tissue.

Five patients with subretinal neovascular complications of angioid streaks were treated with the argon laser. One patient, additionally had a central retinal vein occlusion with preretinal neovascularization. In four out of five cases we obliterated the subretinal neovascular membranes with ensuing improvement of visual acuity.

Bilateral cataract was found in a case of acrodermatitis enteropathica. A possible role of zinc deficiency in causing cataract might be assumed.

The fluoroangiographic examination of the acute stage of retinal lesion in a 16-year-old girl suffering from SSPE showed an exclusively localized retinal vascular involvement 4 days after the onset of her visual complaints. A week later this occlusive vascular lesion progressed to a large area of capillary bed obstruction and it was followed up angiographically. The choroid was involved even during the late stages of evolution of retinal lesion at a low degree.

A report on three patients with illustrations of their visual anomalies following light coagulation for their CSR disease is presented. The correspondence of the phenomena illustrated with the ophthalmoscopically visible fundus changes prove their objective origin.
