Using the rat anti-müllerian hormone type II receptor (AMHRII) cDNA as a probe, two overlapping lambda phage clones containing the AMHRII gene were isolated from a human genomic library. Sequence analysis of the exons was performed and the exon/intron boundaries were determined. The coding region was found to consist of 11 exons, divided over 8 kb.

The acute phase response is a collection of physiologic changes initiated early in the inflammatory process. This response is comprised of both localized changes at the site of infection or injury and the initiation of systemic responses, such as the increase in production of acute phase proteins. Cytokines such as interleukin (IL)-1, IL-6, and tumor necrosis factor (TNF) play key roles in the regulation of acute phase response in the species studied to date.

Comparative genomic hybridization (CGH) was recently developed as a tool to survey entire genomes for variations in DNA sequence copy numbers. We have applied this technique to detect and map amplified regions in 54 soft tissue sarcomas. Aberrations were detected by visual analysis of hybridizations or contrast-enhanced digital images, followed by quantitative digital ratio imaging of the aberrant chromosomes.

Background: Because our laboratory had used the Ciba Corning 200 series blood-gas analyzers for a number of years, we were asked to participate in the evaluation of a premarket unit of the Model 840 analyzer (C840). DESCRIPTION OF DEVICE: The C840 is a bench-top instrument that combines a menu-driven user interface with an automated sampler and integrates software for data management and system diagnostics.

Evaluation Methods: We compared the performance of the C840 to a laboratory-based Ciba Corning 278 (C278), analyzing a total of 325 blood samples.

Terminology in blood pH and gas analysis can be confusing, both because more than one name has been used for the same quantity, and because the same name has been used for more than one quantity. In addition, several calculated quantities are commonly used, but in some cases many different algorithms have been published for a single quantity. This document contains definitions of the most useful quantities in blood pH and gas analysis, and presents algorithms for the most useful calculated quantities.

There has been much attention recently on the working conditions, practice and training of junior doctors in Britain. This article is a personal account of an exchange year spent as a senior house officer in Germany (1993-4). It illustrates differences between the countries and suggests what can be learnt from the experience.

Bovine herpesvirus-1 (BHV-1) is an important pathogen of respiratory infections in cattle. Its continuing importance lies in its ability to predispose infected hosts to bacterial infections (e.g.

A P1 phage clone containing the near complete protein coding region of the human histo-blood group ABO gene locus was isolated and characterized. The insert was estimated at approximately 41 kp, and an approximately 20 kbp BamHI fragment contained the near complete coding region as evaluated by Southern blot analysis. Intron/exon boundaries were determined and the coding region was found to span at least seven exons.

Many doctors view informed consent as merely a medicolegal concept. This article points out that other discourses are possible and that patients would benefit if doctors were to take a wider view.

The relationship between proliferative activity and the expression of pp60c-src in gastric mucosa (oxyntic gland area) of young (4-month) and aged (24-month) Fischer 344 rats was examined. Gastric mucosal proliferative activity, as assessed by 5-bromo-2'-deoxyuridine (BrdU) immunoreactive cells, was found to be 115% (p < .001) higher in aged than in young rats.

We evaluated a new portable instrument, the PPG StatPal II pH and Blood Gas Analysis System, designed for "point-of-care" measurements of blood gases and pH. Inaccuracy (% of target value) and imprecision (CV%) were assessed by blood tonometry and comparison with a Corning 178. Within-day results for PCO2 inaccuracy and imprecision ranged from 98.

We measured total hemoglobin (CtHb) and carboxyhemoglobin (COHb) in 100 patients' blood samples by using five specialized spectrophotometers (CO-oximeters)--IL 482 CO-Oximeter, Corning 2500 CO-oximeter, Radiometer OSM 3 Hemoximeter, Corning 270 CO-oximeter, and the AVL 912 CO-Oxylite--and compared the results with those obtained with the manual cyanmethemoglobin method and a gas-chromatographic (GC) method, respectively. For the CtHb measurements, the differences between the cyanmethemoglobin method and the CO-oximeters were not clinically important for any model. For the blood COHb measurements, the direction of the bias relative to GC was dependent on COHb concentration.

SPRR genes (formerly SPR) encode a novel class of polypeptides (small proline rich proteins) that are strongly induced during differentiation of human epidermal keratinocytes in vitro and in vivo. Recently we found that the N- and C-terminal domains of these proteins show strong sequence homology to loricrin and involucrin, suggesting that SPRR proteins constitute a new class of cornified envelope precursor proteins. Here we show that SPRR proteins are encoded by closely related members of a gene family, consisting of two genes of SPRR1, approximately seven genes for SPRR2, and a single gene for SPRR3.

We evaluated a new commercially available partially purified reduced bovine hemoglobin solution (RBHS) and a new tonometer for use in the quality control of blood gas analyzers. RBHS is manufactured in three different formulations, each corresponding to three different bicarbonate-buffering capacities and concentrations of total hemoglobin (ctHb). The P50 for each formulation of RBHS was determined to be as follows: 30.

The effect of acute or chronic immunoneutralization of somatostatin (SRIF) on plasma GH, thyrotrophin (TSH) and thyroid hormones was examined. Acute responses to SRIF immunoneutralization were examined using 30 intact male lambs (19.8 +/- 0.

A novel approach to the identification of human chromosomes has been developed. Chromosomal in situ hybridization (or "chromosome painting") has been performed using Alu element-mediated PCR products from small quantities (250-500) of flow-sorted normal and abnormal chromosomes. Chromosome paints for various normal chromosomes, including 5, 6, 7, 14, 18, 19, 21, and 22, were generated and shown to be effective in the identification of the appropriate chromosomes.

The efficacy of purified cross-linked human hemoglobin solution in maintaining the metabolic integrity of perfused livers from fed rats was determined and compared with that of livers recirculated with red blood cells, perfluorocarbon, and Krebs' Ringer bicarbonate buffer media at normal and accelerated flow rates. The data indicate that oxygen utilization was comparable in livers perfused with red blood cell medium (53.5 +/- 4.

We have isolated cDNAs encoding the alpha 6 subunit from a lambda gt11 expression library from human keratinocytes by combined screening with a rabbit polyclonal anti-alpha 6 antibody and the polymerase chain reaction. The alpha 6 subunit encoded by this cDNA consists of 1050 amino acids with a 991-amino-acid extracellular, a 23-amino-acid transmembrane and a 36-amino-acid cytoplasmic domain. The extracellular domain contains three putative divalent cation-binding sites and nine potential N-linked glycosylation sites.

Tonometered whole-blood and plasma specimens were tested in plastic and glass syringes to determine whether clinically significant changes in gas tensions occur during sample storage. When whole blood was tonometered with 60 and 100 mL/L (6% and 10%) oxygen and then stored for 30 min in iced plastic syringes, the pO2 of the samples remained stable (mean change = +0.4 and +0.
