Little is known about changes in the abundance of tumor-infiltrating lymphocytes (TILs) and immune phenotype (IP) in recurrent and/or metastatic (R/M) head and neck squamous cell carcinoma (HNSCC). We aimed to compare the TILs and IP between initial and recurrent HNSCCs using paired analysis. Thirty-seven patients who experienced recurrence after surgical resection and received treatment with immune checkpoint inhibitors (ICIs) between June 2014 and June 2023 were included.

Article Synopsis
  • - Abdominal aortic calcification (AAC), a sign of vascular disease, was found to be linked to rapid weight loss in older women, with 39% of studied women experiencing significant weight loss over 5 years.
  • - Women with higher levels of AAC had increased odds of rapid weight loss, even after considering factors like diet and health metrics; this trend was consistent across different dietary and activity subgroups.
  • - Rapid weight loss over 5 years was found to correlate with higher all-cause mortality rates over the subsequent 9.5 years, indicating potential health risks associated with both AAC and weight loss.

Mass spectrometry (MS) based proteomics is widely used for biomarker discovery. However, often, most biomarker candidates from discovery are discarded during the validation processes. Such discrepancies between biomarker discovery and validation are caused by several factors, mainly due to the differences in analytical methodology and experimental conditions.

Article Synopsis
  • New drugs called KRAS inhibitors, like adagrasib and sotorasib, help treat some cancers but sometimes stop working because the cancer cells become resistant.
  • In a study with 38 patients, researchers found that almost half had changes in their cancer cells that allowed them to resist the treatment, with some having more than one change.
  • The study shows that we need better strategies to deal with this resistance to continue helping cancer patients effectively.

Background: Attention-deficit/hyperactivity disorder (ADHD) and body mass index (BMI) are associated. However, it remains unclear whether this association reflects causal relationships in either direction or confounding. Here, we implemented genetically informed methods to examine bidirectional causality and potential confounding.

Article Synopsis
  • There's been a lot of new research on tiny structures called extracellular vesicles (EVs) that cells release, which help us understand how cells work and what goes wrong in diseases.
  • Scientists have had a hard time studying these EVs because they come in different types and can be tough to separate and analyze properly.
  • The International Society for Extracellular Vesicles updated their guidelines, called MISEV2018, to help researchers share clear information about how to study EVs and ensure their findings are accurate and reliable.

Unlabelled: Breast, ovarian, and prostate cancers are hormone-related and may have a shared genetic basis, but this has not been investigated systematically by genome-wide association (GWA) studies. Meta-analyses combining the largest GWA meta-analysis data sets for these cancers totaling 112,349 cases and 116,421 controls of European ancestry, all together and in pairs, identified at P < 10(-8) seven new cross-cancer loci: three associated with susceptibility to all three cancers (rs17041869/2q13/BCL2L11; rs7937840/11q12/INCENP; rs1469713/19p13/GATAD2A), two breast and ovarian cancer risk loci (rs200182588/9q31/SMC2; rs8037137/15q26/RCCD1), and two breast and prostate cancer risk loci (rs5013329/1p34/NSUN4; rs9375701/6q23/L3MBTL3). Index variants in five additional regions previously associated with only one cancer also showed clear association with a second cancer type.

Previous genome-wide association studies have identified common variants in genes associated with bone mineral density (BMD) and risk of fracture. Recently, we identified single nucleotide polymorphisms (SNPs) in Wingless-type mouse mammary tumor virus integration site (WNT)16 that were associated with peak BMD in premenopausal women. To further identify the role of Wnt16 in bone mass regulation, we created transgenic (TG) mice overexpressing human WNT16 in osteoblasts.
