The structural organization of eukaryotic genomes is contingent upon the fractionation of DNA into transcriptionally permissive euchromatin and repressive heterochromatin. However, we have a limited understanding of how these distinct states are first established during animal embryogenesis. Histone 3 lysine 9 trimethylation (H3K9me3) is critical to heterochromatin formation, and bulk establishment of this mark is thought to help drive large-scale remodeling of an initially naive chromatin state during animal embryogenesis.

Article Synopsis
  • TAT (Trans-activating transcriptional activator) is a peptide that helps drugs enter cells and target the nucleus, but it has issues with blood circulation due to interactions with serum components.
  • The study developed a new nanoplatform, DA-TAT-PECL, to reduce TAT's undesired interactions by modifying it with carboxylic acid and combining it with a polymer, allowing for prolonged circulation and targeted tumor delivery.
  • Additionally, an anticancer drug, camptothecin (CPT), was modified to monitor its release using a specific bond, demonstrating the effectiveness of this new pH/reduction-responsive micelle system for cancer therapy.

Endothelial dysfunction and blood-brain barrier (BBB) leakage have been suggested as a fundamental role in the development of cerebral small vessel disease (SVD) pathology. However, the molecular and cellular mechanisms that link cerebral hypoxic hypoperfusion and BBB disruption remain elusive. Sphingosine-1-phosphate (S1P) regulates the BBB integrity by binding to its receptor isoform 1 (S1PR1) on endothelial cells.

Background: Glioblastoma is the most common primary malignant and treatment-resistant human brain tumor. Rodent models have played an important role in understanding brain cancer biology and treatment. However, due to their small cranium and tumor volume mismatch, relative to human disease, they have been less useful for translational studies.
