We conducted a phase Ib/II clinical trial to evaluate the safety, feasibility, and clinical activity of combining pembrolizumab (anti-PD-1) with XL888 (Hsp90 inhibitor) in patients with advanced colorectal cancer (CRC). We hypothesized that this regimen would modulate soluble and cellular immune mediators and enhance clinical outcomes. The trial employed a 3 + 3 open-label design, with an expansion cohort at the recommended phase II dose (RP2D) in treatment-refractory, mismatch repair-proficient CRC patients.

Little is known regarding the role of CPAP therapy in reversing the risk of AF recurrence in patients with OSA. Awareness of the therapeutic effects of CPAP in preventing AF recurrence post-ablation will help create a multidisciplinary team of doctors to treat this complex population of patients.

A commonly used strategy for the extension of protein half-life in circulation is the introduction of negative charges on the protein, preferably through N- and O-glycans equipped with negatively charged sialic acid moieties. While enhancing pharmacokinetics, the presence of many glycosylation sites can coincide with a considerable protein heterogeneity, which renders analytical characterisation increasingly difficult.
