Radiotherapy, employing high-energy rays to precisely target and eradicate tumor cells, plays a pivotal role in the treatment of various malignancies. Despite its therapeutic potential, the effectiveness of radiotherapy is hindered by the tumor's inherent low radiosensitivity and the immunosuppressive microenvironment. Here we present an innovative approach that integrates peroxynitrite (ONOO)-mediated radiosensitization with the tumor-associated neutrophils (TANs) polarization for the reversal of immunosuppressive tumor microenvironment (TME), greatly amplifying the potency of radiotherapy.

Controlling CRISPR/Cas9 gene editing at the spatiotemporal resolution level, especially for in vivo applications, remains a great challenge. Here, we developed a near-infrared (NIR) light-activated nanophotonic system (UCPP) for controlled CRISPR-Cas9 gene editing and synergistic photodynamic therapy (PDT). Lanthanide-doped upconversion nanoparticles are not only employed as carriers for intracellular plasmid delivery but also serve as the nanotransducers to convert NIR light (980 nm) into visible light with emission at 460 and 650 nm, which could result in simultaneous activation of gene editing and PDT processes, respectively.

Metal-semiconductor nanozyme of dumbbell Au-CeO with spatially separated heterostructure has cascade superoxide dismutase (SOD)-like and peroxidase (POD)-like activities for superoxide anions detection. It was synthesized by selective growth of CeO at the ends of Au nanorod (Au NR). Taking advantage of the excellent local surface plasmon resonance (LSPR) effect of Au NR, the spatially separated Au-CeO has a higher photothermal effect than the continuously growing core-shell structure of Au@CeO.

A novel construction strategy is introduced for an ultrasensitive dynamic light scattering (DLS) immunosensor targeting alpha fetoprotein (AFP). This approach relies on a self-assembled heptamer fusion protein (A1-C4bpα), incorporating the dual functions of multivalent recognition and crosslinking aggregation amplification due to the presence of seven AFP-specific A1 nanobodies on the A1-C4bpα heptamer. Leveraging antibody-functionalized magnetic nanoparticles for target AFP capture and DLS signal output, the proposed heptamer-assisted DLS immunosensor offers high sensitivity, strong specificity, and ease of operation.

AlCl-mediated nucleophilic ring-opening reactions of indoline-2-thiones with various acyl cyclopropanes, bi-cyclopropanes and spirocyclic cyclopropanes were investigated. A series of ketones functionalized with indolylthio groups were synthesized in yields ranging from moderate to good. Moreover, chemical transformations of 4-indolylthio butan-1-ones to dihydro-2-thiepino[2,3-]indoles and sulfone were carried out to further expand both synthetic utility and structural complexity.

Traditional oxidase-like (OXD) nanozymes rely primarily on O-mediated superoxide anion (O) process for catalytic oxidation and organophosphorus (Ops) detection. While during the actual detection process, the concentration of O is inconstant that can be easily changed with the external environment, distorting detection results. Herein, highly-oxidizing Au@MnO nanozymes with core-shell nanostructure are designed which trigger substantial electron transfer from inner Au core to outer ultrathin MnO layer.

Cardiac fibrosis, a common pathology in inherited and acquired heart diseases, necessitates the identification of diagnostic and therapeutic targets. Methyltransferase Like 1 (METTL1), an enzyme responsible for RNA modification by methylating guanosine to form m7G, is an emerging area of research in understanding cellular processes and disease pathogenesis. Dysregulation of m7G modification has been implicated in various diseases.

Article Synopsis
  • - Most enzyme catalysts struggle with oxidation resistance due to their limited mimicking abilities and the generation of harmful reactive oxygen species (ROS).
  • - Researchers created a novel Au@CuO heterostructure that demonstrates improved antioxidant capacity, effectively scavenging damaging secondary ROS like hydroxyl radicals (·OH).
  • - The heterostructure functions like various antioxidant enzymes and can eliminate ·OH under both acidic and neutral conditions, showcasing its potential in real-world applications like A549 cells and zebrafish.

The traditional quantitative analysis methods of ascorbic acid (AA), which require expensive equipment, a large amount of samples and professional technicians, are usually complex and time-consuming. A low-cost and high-efficiency AA detection device is reported in this work. It integrates a three-electrode sensor module prepared by screen printing technology, and a microfluidic chip with a finger-actuated micropump peeled from the liquid-crystal display (LCD) 3D printing resin molds.

Objectives: Serum neurofilament light chain (NfL) is a biomarker for neuroaxonal damage, and S100B is a blood marker for cerebral damage. In the present study, we investigated the relationship between serum NfL and S100B levels, severity, and outcomes in patients with aneurysmal subarachnoid hemorrhage (aSAH).

Methods: We prospectively recruited aSAH patients and healthy controls between January 2016 and January 2021.

Generally, a high quality surface-enhanced Raman spectroscopy (SERS) substrate often requires a highly-tailorable electromagnetic (EM) field generated at nanoparticle (NP) surfaces by the regulation of the morphologies, components and roughness of NPs. However, most recent universal approaches are restricted to single components, and integrating these key factors into one system to achieve the theoretically maximum signal amplification is still challenging. Herein, we design a triple SERS signal amplification platform by the coordination of spiky Au NPs with rich-tip nanostructures, controllable silver nanoshell, as well as tailorable surface roughness into one nano-system.

Oxidative stress is considered an important pathogenic process of cardiac hypertrophy. Lycopene is a kind of carotenoid antioxidant that protects the cardiovascular system, so we hypothesized that lycopene might inhibit cardiac hypertrophy by attenuating oxidative stress. Phenylephrine and pressure overload were used to set up the hypertrophic models in vitro and in vivo respectively.

Background: The bone marrow microenvironment constitutes a sanctuary for leukemia cells. Recent evidence indicates that environment-mediated drug resistance arises from a reciprocal influence between tumor cells and the surrounding stroma. The present study aimed to investigate the effect of chronic lymphocytic leukemia (CLL) cells on the metabolism of bone marrow stroma, to determine the role of this metabolic change in the stroma in vorinostat resistance of CLL cells, and thus to assess a novel strategy to target stroma and achieve the maximum therapeutic effect of vorinostat.
