Immune checkpoints are crucial for regulating immune responses and maintaining self-tolerance, as they play a pivotal role in preventing autoimmunity and facilitating tumor immune evasion. This review concentrates on the immune checkpoint molecules PSGL-1 and VISTA. Both molecules are highly expressed in hematopoietic cells, including T cells and myeloid cells.

Article Synopsis
  • - The study aimed to predict the prognosis of antibody-positive autoimmune encephalitis (AE) patients using nine different machine learning methods, analyzing data from 187 patients divided into training (121) and validation (67) sets.
  • - Among the machine learning models evaluated, random forest (RF) achieved the highest accuracy (0.860), followed by extreme gradient boosting (XGBoost) with an accuracy of 0.826, demonstrating their effectiveness in predicting patient outcomes.
  • - Despite the promising results, the study acknowledges a limitation due to the small sample size typical for a rare disease, but concludes that the RF and XGBoost models exhibit strong performance in terms of discrimination, calibration, and clinical applicability.

Article Synopsis
  • Machine learning (ML) techniques were applied to predict the prognosis of Guillain-Barré syndrome (GBS) by analyzing medical records from 223 patients and constructing various predictive models.
  • Eight ML algorithms, including XGBoost, Random Forest, and Light Gradient Boosting Machine, were evaluated, with XGBoost showing the highest accuracy and F1 score.
  • Key features impacting prognosis included blood neutrophil/lymphocyte ratio, age, and mechanical ventilation, indicating these factors play a significant role in short-term outcomes for GBS patients.

Objectives: To investigate the risk factors of pulmonary infection in patients with severe myelitis and construct a prediction model.

Methods: The clinical data of 177 patients with severe myelitis at admission from the First Affiliated Hospital of Zhengzhou University from January 2020 to December 2022 were retrospectively analyzed. The predicting factors associated with pulmonary infection were screened by multivariate logistic regression analysis, and the nomogram model was constructed, and the predictive efficiency of the model was evaluated, which was verified by calibration curve, Hosmer-Lemeshow goodness-of-fit test and decision curve analysis.

LILRB4, a myeloid inhibitory receptor belonging to the family of leukocyte immunoglobulin-like receptors (LILRs/LIRs), plays a pivotal role in the regulation of immune tolerance. LILRB4 primarily mediates suppressive immune responses by transmitting inhibitory signals through immunoreceptor tyrosine-based inhibitory motifs (ITIMs). This immune checkpoint molecule has gained considerable attention due to its potent regulatory functions.

Major histocompatibility complex class I related chain A (MICA) is an important and stress-induced ligand of the natural killer group 2 member D receptor (NKG2D) that is expressed in various tumour cells. Given that the MICA/NKG2D signalling system is critically embedded in the innate and adaptive immune responses, it is particularly involved in the surveillance of cancer and viral infections. Emerging evidence has revealed the important roles of non-coding RNAs (ncRNAs) including microRNAs (miRNAs), long noncoding RNAs (lncRNAs) and circular RNAs (circRNAs) in different cancer types.

Objective: The purpose of this research was to evaluate the influence of immunity on infection in patients with severe hemorrhagic stroke and explore the mechanism underlying this connection.

Methods: Clinical data obtained from 126 patients with severe hemorrhagic stroke were retrospectively analyzed, and the factors affecting infection were screened by multivariable logistic regression models. Nomograms, calibration curves, the Hosmer-Lemeshow goodness-of-fit test, and decision curve analysis were used to examine the effectiveness of the models in evaluating infection.

Article Synopsis
  • * Researchers analyzed a total of 2,182 NGS results from 1,484 patients, finding HPV B19 in 39 samples from 33 patients, predominantly in cerebrospinal fluid and blood.
  • * The majority of those infected had underlying conditions that lowered their immunity and presented with symptoms like fever, anemia, and severe nervous system issues, with a significant number improving after antiviral treatment.

A severely comatose female patient was diagnosed with Japanese encephalitis (JE). Her condition was complicated by Hashimoto's thyroiditis (HT) and Guillain-Barré syndrome (GBS). After antiviral, glucocorticoid, and immunoglobulin treatment, the patient's consciousness was restored, and she could breathe spontaneously.

There is currently no effective treatment for Japanese encephalitis, which has a high rate of morbidity and mortality. This study assessed the effectiveness of a ganciclovir, methylprednisolone, and immunoglobulin combination (TAGMIC) therapy in decreasing cognitive impairment and mortality among patients with Japanese encephalitis. We retrospectively assessed the clinical data of 31 patients diagnosed with Japanese encephalitis, who were admitted to an intensive care unit.
