Background: Neoadjuvant chemotherapy (NACT) has arisen as a treatment option for breast cancer (BC). However, the response to NACT is still unpredictable and dependent on cancer subtype. Metabolomics is a tool for predicting biomarkers and chemotherapy response.

Introduction: Studies addressing the methylation pattern in adamantinomatous craniopharyngioma (ACP) are lacking.

Objective: To identify methylation signatures in ACPs regarding clinical presentation and outcome.

Methods: Clinical and pathology data were collected from 35 patients with ACP (54% male; 18.

Objective: To explore pituitary tumors by methylome and transcriptome signatures in a heterogeneous ethnic population.

Methods: In this retrospective cross-sectional study, clinicopathological features, methylome, and transcriptome were evaluated in pituitary tumors from 77 patients (61% women, age 12-72 years) followed due to functioning (FPT: GH-secreting n = 18, ACTH-secreting n = 14) and nonfunctioning pituitary tumors (NFPT, n = 45) at Ribeirao Preto Medical School, University of São Paulo.

Results: Unsupervised hierarchical clustering analysis (UHCA) of methylome (n = 77) and transcriptome (n = 65 out of 77) revealed 3 clusters each: one enriched by FPT, one by NFPT, and a third by ACTH-secreting and NFPT.

The COVID-19 pandemic boosted the development of diagnostic tests to meet patient needs and provide accurate, sensitive, and fast disease detection. Despite rapid advancements, limitations related to turnaround time, varying performance metrics due to different sampling sites, illness duration, co-infections, and the need for particular reagents still exist. As an alternative diagnostic test, we present urine analysis through flow-injection-tandem mass spectrometry (FIA-MS/MS) as a powerful approach for COVID-19 diagnosis, targeting the detection of amino acids and acylcarnitines.

Neoadjuvant chemotherapy (NACT) is offered to patients with operable or inoperable breast cancer (BC) to downstage the disease. Clinical responses to NACT may vary depending on a few known clinical and biological features, but the diversity of responses to NACT is not fully understood. In this study, 80 women had their metabolite profiles of pre-treatment sera analyzed for potential NACT response biomarker candidates in combination with immunohistochemical parameters using Nuclear Magnetic Resonance (NMR).

Article Synopsis
  • The study aims to assess the therapeutic potential of vitamin D receptor (VDR) signaling in adrenocortical carcinoma (ACC) cells, specifically H295R.
  • Researchers found that VDR was hypermethylated and underexpressed in these cells, but treatments with calcitriol and seocalcitol restored VDR signaling, reduced cell proliferation, and affected other signaling pathways.
  • In animal models, seocalcitol inhibited tumor growth and reduced steroid secretion from tumors without severe side effects, suggesting VDR could be a promising target for ACC therapy and patient management.

Children diagnosed with pediatric adrenocortical tumors (pACT) have variable outcomes, and, to date, the disease lacks robust prognostic biomarkers. The prognostic potential of tumor methylation has been demonstrated in several cancers. We aimed to evaluate the pACT methylation profile and its association with disease presentation and survival.

Article Synopsis
  • Pediatric adrenocortical tumors (pACT) show complex genomic profiles and lack clear prognostic markers and treatment options, prompting research into the Vitamin D3 receptor (VDR) for potential insights.
  • A study involving 108 pediatric patients found that most pACT had low VDR expression and high methylation, particularly in more aggressive carcinomas, suggesting a link between VDR status and disease severity.
  • The findings indicate that VDR hypermethylation and underexpression could serve as important predictive and prognostic biomarkers for pACT, affecting patient outcomes.

The outbreak of COVID-19 has created an unprecedent global crisis. While the polymerase chain reaction (PCR) is the gold standard method for detecting active SARS-CoV-2 infection, alternative high-throughput diagnostic tests are of a significant value to meet universal testing demands. Here, we describe a new design of the MasSpec Pen technology integrated to electrospray ionization (ESI) for direct analysis of clinical swabs and investigate its use for COVID-19 screening.

Metacaspases are cysteine proteases present in plants, fungi, prokaryotes, and early branching eukaryotes, although a detailed description of their cellular function remains unclear. Currently, three-dimensional (3D) structures are only available for two metacaspases: Trypanosoma brucei (MCA2) and Saccharomyces cerevisiae (Yca1). Furthermore, metacaspases diverged from animal caspases of known structure, which limits straightforward homology-based interpretation of functional data.
