Members of the genus including Jurona virus (JURV) have emerged as promising immunotherapeutic agents, characterized by their tumor selectivity, fast kinetics, low seroprevalence, and minimal toxicity in humans. Here, we demonstrate that the administration of JURV leads to tumor regression in both hepatocellular carcinoma (HCC) xenograft and syngeneic models. Furthermore, our findings indicate that combining JURV and anti-PD-1 therapy reduced tumor burden and improved survival rates over JURV or anti-PD-1 alone in an orthotopic HCC model.

A novel Cu/Pd-catalyzed domino radical cyclization and C-H carbonylation of α-bromocarbonyls has been developed, which enables the rapid incorporation of CO unit into polycyclic quinolin-2(1)-one scaffolds. By using Mo(CO) as the CO source, the reaction proceeded smoothly to furnish various polycyclic carbonyl-containing quinolin-2(1)-one derivatives in high yields. Notably, this method could be used in the late-stage modification of biologically active molecules such as estrone.

With the aging process of the global population and the development of medical technology, the cases of postoperative cognitive dysfunction (POCD) are also increasing. Due to the complexity of the pathogenesis, urgent treatment has been sought. Neuroinflammation induced by the accumulation of lipid droplets (LDs) in microglia has been closely watched in recent years and is also considered to be an important cause of nerve damage.

Article Synopsis
  • - This study explored how mitochondrial dynamics influence postoperative cognitive dysfunction (POCD) and tested the drug Mdivi-1, which inhibits a protein called DRP1, as a potential treatment.
  • - Results showed that inhibiting DRP1 with Mdivi-1 reduced neuroinflammation and improved cognitive function by decreasing mitochondrial fission and reactive oxygen species (ROS) production, while also stabilizing the mitochondrial membrane.
  • - Additionally, Mdivi-1 increased synaptic plasticity and spine density in the hippocampus, leading to better cognitive performance in tests, highlighting mitochondrial health as a key factor in POCD and the promise of DRP1 inhibition for treatment.

ARID1A is a key component of the switch/sucrose non-fermentable (SWI/SNF) chromatin remodeling complex and functions as a critical tumor suppressor in various cancers. In this study, we find that tumor cells with hotspot missense mutations in ARID1A (AT-rich interactive domain-containing protein 1A) exhibit a malignant phenotype. Mechanistically, these mutations facilitate the translocation of ARID1A mutant proteins to the cytoplasm by the nucleocytoplasmic shuttler XPO1 (exportin 1).

Style penetration by pollen tubes is essential for reproductive success, a process requiring canonical Rab5s in Arabidopsis. However, functional loss of Arabidopsis Vps9a, the gene encoding for guanine nucleotide exchange factor (GEF) of Rab5s, did not affect male transmission, implying the presence of a compensation program or redundancy. By combining genetic, cytological, and molecular approaches, we report that Arabidopsis Vps9b is a pollen-preferential gene, redundantly mediating pollen tube penetration of style with Vps9a.
