Background: Breast Cancer (BC) is the most common type of cancer in women around the world and 70% of cases are hormone-receptor positive (HR+). In 40% of cases, a key mechanism of endocrine resistance to the standard first line is a mutation of the ligand-binding domain (LBD) of Estrogen Receptor 1 (ESR1) encoding estrogen receptor α (ER). Most common ESR1 mutations that occur at positions 537 and 538 have been associated with poor clinical outcomes.

Tissue-resident memory (T) T cells have emerged as key players in cancer immunosurveillance, and their presence has been linked to a favorable clinical outcome in solid cancer patients. Liver metastases exhibit a highly immunosuppressive tumor microenvironment, however, the role and clinical impact of T cell infiltration in colorectal cancer remain elusive. The expression of several tissue residency and activation biomarkers has been investigated on tumor-infiltrating lymphocytes isolated from 26 patients' colorectal cancer liver metastases (CRC liver metastases) and compared to 16 peripheral blood samples of patients with CRC liver metastases.

Background: Although the benefit of supportive care in the postcancer period is now well demonstrated, its implementation in the patient journey remains challenging. This article describes the development, since 2015 and in routine care, of supportive postcancer care comprising a multidisciplinary rehabilitation programme (MRP) based on exercise for patients with early breast cancer.

Methods: As part of quality control, we reviewed all patient files since the programme was implemented.

Article Synopsis
  • Colorectal cancer is a major health concern, ranking as the third leading cause of cancer deaths globally in 2022, and early detection through screening is crucial for improving outcomes.
  • Using bioinformatics, researchers identified new DNA methylation biomarkers (COL25A1 and METAP1D) for colorectal cancer by analyzing extensive genetic data, which was then validated through specific testing on samples from patients and healthy individuals.
  • The biomarkers showed high specificity (100%) and moderate sensitivity (49%) for detecting colorectal cancer in blood samples, suggesting their potential for noninvasive screening, though further studies are needed to confirm their practical use in clinical settings.

  • K-mer indexing is an effective technique for searching RNA sequences in RNA-seq libraries but hasn't allowed for direct quantification of RNA until now.
  • The study demonstrates that arbitrary RNA sequences can be quantified quickly and accurately by breaking them down into k-mers, achieving precision similar to traditional methods.
  • Using an extensive RNA-seq sample collection from the Cancer Cell Line Encyclopedia, the researchers showcase how k-mer indexing can uncover non-reference sequences and variant RNAs related to specific genetic changes.

Article Synopsis
  • Immune checkpoint inhibitors (ICIs) are a crucial treatment for various cancers, but grade≥3 immune-related adverse effects (irAEs) are not well studied; this research aimed to evaluate their incidence and characteristics.
  • The study reviewed patients with advanced melanoma, lung cancer, or renal cancer who experienced severe irAEs while undergoing ICI therapy from 2016-2021, revealing a grade≥3 irAE incidence of 11.7%.
  • The majority of severe adverse effects were hepatobiliary, gastrointestinal, musculoskeletal, and respiratory, typically occurring around 6 months into treatment, with a significant percentage leading to hospitalization; however, a majority of patients recovered without lasting issues.

Context And Aims: Eribulin is used in taxane and anthracycline refractory HER2-negative metastatic breast cancers (MBC). Patients treated in pivotal clinical trials achieved low survival rates, therefore, the identification of prognostic criteria for long progression-free survival (PFS) is still an unmet medical need. In this study, we sought to determine potential prognostic criteria for long-term eribulin response in HER2-negative MBC.

Background: Up to 70% of breast cancer patients report symptoms of insomnia during and after treatment. Despite the ubiquity of insomnia symptoms, they are under-screened, under-diagnosed and poorly managed in breast cancer patients. Sleep medications treat symptoms but are ineffective to cure insomnia.

Unlabelled: Metastatic relapse after treatment is the leading cause of cancer mortality, and known resistance mechanisms are missing for most treatments administered to patients. To bridge this gap, we analyze a pan-cancer cohort (META-PRISM) of 1,031 refractory metastatic tumors profiled via whole-exome and transcriptome sequencing. META-PRISM tumors, particularly prostate, bladder, and pancreatic types, displayed the most transformed genomes compared with primary untreated tumors.

CD226 has been reported to participate in the rescue of CD8 T cell dysfunction. In this study, we aimed to assess the prognostic value of CD226 in tumor-infiltrating lymphocytes (TILs) derived from colorectal cancer (CRC) liver metastases treated with chemotherapy and radical surgery. TILs from 43 metastases were isolated and analyzed ex vivo using flow cytometry.

Article Synopsis
  • Combining immunogenic chemotherapy (FOLFOX) with anti-PD-1 can significantly enhance tumor responses, but TGF-β1 signaling remains a barrier to effective treatment.
  • Research using mouse models revealed that blocking TGF-β1 enhances the effectiveness of this combination therapy by recruiting more CD8 T cells to the tumor.
  • TGF-β1's immune-suppressive effects are linked to epigenetic changes in cancer-associated fibroblasts, which hinder the production of important chemokines (CXCL9/10) necessary for a robust immune response, thus limiting the success of chemo-immunotherapy.

Article Synopsis
  • - High levels of Ki67 in breast cancers correlate with poor outcomes, and the study focused on the agreement between Ki67 levels from the Oncotype DX test and standard Ki67 measurements in early-stage ER+/HER2- breast cancers.
  • - The study found a low correlation (0.288) between Ki67 values from Oncotype DX and traditional lab methods, with Ki67 levels being significantly linked to event occurrences (p = 0.03) but not to event-free survival (EFS) (p = 0.26).
  • - The results suggest that while there’s a disconnect in Ki67 measurements, the gene may still hold prognostic potential, especially in regions lacking access to Oncotype DX

Background: The positive role of CD8+ tumor-infiltrating lymphocytes (TIL) in patients with colorectal cancer (CRC) has been well described but the prognostic value of CD4 T cell subsets remained to be investigated. In this study, we expanded TIL from surgically resected liver metastases of patients with CRC and characterized the phenotype and the prognostic value of expanded-CD4 T cells.

Methods: Liver metastases were surgically resected from 23 patients with CRC.

Angiopoietin-2 (ANGPT2) is a prognostic factor in metastatic colorectal cancer (CRC). Nevertheless, it remains to be elucidated which molecular characteristics make up the ANGPT2-related poor-prognosis CRC subset. Public transcriptomic datasets were collected from Gene Expression Omnibus GEO and with the TCGAbiolinks R-package for the TCGA.

We conducted a retrospective study to assess the follow-up of patients with localized breast cancer and the first indicators of advanced breast cancer recurrence. All patients with advanced breast cancer recurrence treated between January 2010 and June 2016 in our institution were registered. Among these patients, 303 patients initially treated for early breast cancer with curative intent were identified.

Unlabelled: In France, HIV-infected (HIV+) patients are frequently coinfected with hepatitis B virus (HBV) or at risk for this infection. Physicians and their patients should be more committed to HBV prevention than the average population.

Aims: To gain insight into the attitude towards HBV and its vaccination in HIV+ patients from the Aquitaine Cohort and their attending physicians in France.
