Introduction: Interleukin-6 (IL-6) is an important mediator of inflammation. In addition to cells involved in inflammation, sensory nociceptive neurons express the IL-6 signal-transducer glycoprotein 130 (gp130). These neurons are not only involved in pain generation but also produce neurogenic inflammation by release of neuropeptides such as calcitonin gene-related peptide (CGRP).

Objective: In arthritis, macrophages invade the affected joint. Experimental arthritis models have shown that macrophages also invade the dorsal root ganglia (DRGs) of the inflamed segments in which the perikarya of sensory neurons are located. It is unclear whether this macrophage invasion contributes to arthritis pain and/or furthers neuronal damage.

Chlamydiae are obligate intracellular pathogens which secrete host-interactive proteins capable of directly modulating eukaryotic pathways. Using the PDZ domain of the protease CT441 of Chlamydia trachomatis as a bait in a yeast two-hybrid screen, we identified the SRAP1 co-activator of estrogen receptor alpha (ERalpha) as an interacting protein. SRAP1 is a unique modulator of steroid receptor activity, as it is able to mediate its co-regulatory effects both as a RNA and a protein.

Lipid homeostasis is essential for proper function of cells and organisms. To unravel new regulators of this system, we developed a screening procedure, combining RNA interference in HeLa cells and TLC, which enabled us to monitor modifications of lipid composition resulting from short, interfering RNA knock-downs. We applied this technique to the analysis of 600 human kinases.
