The assessment of major depressive disorder (MDD) treatment efficacy typically relies on clinician-rated scales of depressive symptoms with a notable absence of predictive biomarkers. In this study, a concentration ratio of serum brain-derived neurotrophic factor (BDNF) to interleukin-6 (IL-6) was identified as a promising predictive biomarker. To overcome the significant disparity in blood levels between BDNF (ng·mL range) and IL-6 (pg·mL range), a dual-pathway signal amplification method involving small carbon dots (Fe-CDs and Pb-CDs) was employed.

Article Synopsis
  • On-surface molecular self-assembly is key for creating low-dimensional functional nanostructures, but traditional methods mainly use hydrogen bonding or metal coordination strategies.
  • This research examines using tellurium (Te)-directed chalcogen bonding for building molecular networks with carbonitrile molecules, achieving unique 2D structures on gold surfaces.
  • Although the Te-carbonitrile interactions are less stable than traditional metal-organic bonds, this approach could broaden techniques for supramolecular assembly and enhance the design of advanced nanomaterials.

Low-mass soluble β-amyloid peptide oligomers (LSAβOs) play a crucial role in the pathogenesis of Alzheimer's disease. However, these oligomers exhibit heterogeneity in terms of structure, stability, and stoichiometry, and their abundance in biofluids is low, making accurate identification challenging. In this study, we developed a DNA nanocage-assisted method for selective sizing and sensitive quantification of LSAβOs in serum.

Major depressive disorder (MDD) is a prevalent brain disorder affecting more than 2% of the world's population. Due to the lack of well-specific biomarkers, it is difficult to distinguish MDD from other diseases with similar clinical symptoms (such as Alzheimer's disease and cerebral thrombosis). In this work, we provided a strategy to address this issue by constructing a combinatorial biomarker of serum glial fibrillary acidic protein (GFAP) and neurofilament light chain (NFL).

Nanoplastics, widely existing in the environment and organisms, have been proven to cross the blood-brain barrier, increasing the incidence of neurodegenerative diseases like Alzheimer's disease (AD). However, current studies mainly focus on the neurotoxicity of nanoplastics themselves, neglecting their synergistic effects with other biomolecules and the resulting neurotoxicity. Amyloid β peptide (Aβ), which triggers neurotoxicity through its self-aggregation, is the paramount pathogenic protein in AD.

The relative positions of viral DNA genomes to the host intranuclear environment play critical roles in determining virus fate. Recent advances in the application of chromosome conformation capture-based sequencing analysis (3 C technologies) have revealed valuable aspects of the spatiotemporal interplay of viral genomes with host chromosomes. However, to elucidate the causal relationship between the subnuclear localization of viral genomes and the pathogenic outcome of an infection, manipulative tools are needed.

The abundance of brain-derived neurotrophic factor (BDNF) in aqueous humor (AH) is an ideal biomarker for the diagnosis of glaucoma, a chronic progressive optic neuropathy and the most frequent cause of irreversible blindness. The difficulty of AH-based BDNF detection is from the small amount of extracted AH in a paracentesis (<100 μL) and the ultra-low abundance of BDNF. In this work, we systematically studied the non-specific adsorption of biofluids on the bare gold electrode by electrochemistry and Raman spectroscopy techniques, revealing the unexpected negative correlation of the extent of non-specific adsorption with the size of the electrode.

Serum neurofilament light chain (NFL), a potential general biomarker for neurodegenerative diseases, is not specific enough to differentiate neurodegenerative diseases from other brain diseases such as cerebral thrombosis (CT). According to the importance of glycosylation in neurodegenerative pathogenesis, the NFL glycosylation level (oNFL/tNFL), defined as the ratio of glycosylated NFL (oNFL) to total NFL (tNFL), may be a more effective index. The major challenge in serum oNFL/tNFL detection is the ultra-low abundance of both NFL forms.

Aptamers are widely used as a recognition element for Alzheimer's disease (AD)-related biomarker determinations. Due to the existing Cu in real samples, and its possible coordinations with aptamers and biomarkers, the aptamer-based biomarker determination could be seriously disturbed. In this work, taking Aβ oligomer (AβO, a typical biomarker for AD) as a model, the interference of Cu in the interaction between AβO and its aptamer (Apt) was investigated by surface plasmon resonance spectroscopy.
