Sepsis is a critical condition with high mortality, often leading to acute lung injury (ALI) due to uncontrolled inflammatory responses and alveolar epithelial damage. Extracellular vesicles (EVs), particularly mesenchymal stem cell-derived EVs, have shown therapeutic potential in sepsis-related organ dysfunction by transferring RNAs and proteins. However, their clinical use is limited by low efficacy and yield.

The significance of glycans in various biological processes has been widely acknowledged. Quantitative glycomics is emerging as an important addition to clinical biomarker discovery, as it helps uncover disease-associated glycosylation patterns that are valuable for diagnosis, prognosis, and treatment evaluation. Compared to glycoproteomics and other established omics approaches, quantitative glycomics exhibits greater methodological diversity and it encounters various challenges in automation and standardization.

A major challenge in structural glycomics is the presence of isomeric glycan structures, which may not be fully resolved by separation techniques such as liquid chromatography (LC) and ion mobility spectrometry (IMS). Tandem mass spectrometry (MS/MS) can be employed following on-line separation to distinguish unresolved features, as the temporal profiles of various fragment ions reflect different combinations of those from their respective precursor ions. However, traditional principal component analysis can produce negative signals that are unrealistic for real data, and classic non-negative matrix factorization (NMF) methods may result in factors that include contributions from multiple components.

Drug resistance has emerged as a critical challenge in clinical cancer treatment. Here, we present a high-throughput screening protocol to identify therapeutic small-molecule inhibitors against drug-resistant cancer cells. We detail the steps for constructing drug-resistant cell models, executing the chemical screening process, and performing data analysis and validation.

In the mammalian cardiovascular system, endothelial glycocalyx is a gel-like layer that covers the luminal surface of endothelial cells (ECs) and plays crucial roles in vascular homeostasis, permeability and leukocyte adhesion. Degradation of this structure occurs early in sepsis and becomes accordingly dysfunctional. In severe cases, it is not self-regulated by the organism.

Background: In gastric cancer (GC) patients, malnutrition is common and has a negative impact on treatment tolerance, survival, and prognosis. The purpose of this study was to explore the relationship between prechemotherapy nutritional state and early myelosuppression in stage IV GC patients treated with first-line chemotherapy.

Methods: This retrospective study included patients with stage IV GC who received first-line chemotherapy between July 2012 and December 2021.

The Arabidopsis transcription factor ATAF1 negatively regulates thermomorphogenesis by inhibiting the expression of key genes involved in thermoresponsive elongation. DET1-mediated ubiquitination promotes ATAF1 degradation. In response to warmer, non-stressful average temperatures, plants have evolved an adaptive morphologic response called thermomorphogenesis to increase their fitness.

Article Synopsis
  • Pyridaben is a commonly used contact acaricide that negatively affects the hunting abilities of predatory mites, particularly by impairing their ability to detect prey through chemical cues.
  • The study showed that exposure to pyridaben reduced key metrics of predation, including efficiency and searching behavior, with effects worsening at higher doses.
  • The downregulation of a specific gene related to olfaction was observed after treatment, indicating that pyridaben disrupts the predatory functions of these mites by interfering with their ability to sense and capture prey.

Controlling multicolor persistent room-temperature phosphorescence (RTP) through photoirradiation holds fundamental significance but remains a significant challenge. In this study, we engineered a wavelength-selective photoresponsive system utilizing the Förster resonance energy transfer strategy. This system integrates a photoactivated long-lived luminescent material as the energy donor with a fluorescent photoswitch as the energy acceptor, facilitating programmable persistent luminescence switches.

Paclitaxel-resistant triple negative breast cancer (TNBC) remains one of the most challenging breast cancers to treat. Here, using an epigenetic chemical probe screen, we uncover an acquired vulnerability of paclitaxel-resistant TNBC cells to protein arginine methyltransferases (PRMTs) inhibition. Analysis of cell lines and in-house clinical samples demonstrates that resistant cells evade paclitaxel killing through stabilizing mitotic chromatin assembly.

The oxygen level in the tumor microenvironment (TME) plays a critical role in regulating cell fates such as proliferation, migration, apoptosis, and so forth. To better elucidate how hypoxia affects tumor cell behaviors, a series of microfluidic strategies have been utilized to generate an oxygen gradient covering both hypoxia and normoxia conditions. However, in most studies, some chemicals are introduced into microfluidic chips, causing the potential of their poor biocompatibility.

Meeting the power demand from the transmission system operator is an important objective for power dispatch, which introduces a power supply-demand equality constraint coupling all the wind turbines among the wind farm into the optimization problem. For a large-scale wind farm, processing the global equality constraint in a centralized or distributed framework is time-consuming and computationally complex. Here we considered the fast and localized execution issue of the power optimal dispatch problems.

Nodal line semimetal (NLSM) has become a captivating medium for studying varieties of novel quantum phenomena. Here, based on first-principles calculations, we identify a square compound lattice (SCL) structure, namely C-Me-graphene, featuring a NLSM, wherein the nodal line of this configuration resides precisely at the Fermi energy without any extraneous bands in the vicinity, manifesting the quintessential characteristics of an ideal NLSM. As a corollary, utilizing symmetry analysis, we propose that nodal lines can be generated by exploiting the two-dimensional (2D) SCL of carbon.
