Immune checkpoint inhibition has shown success in treating metastatic cutaneous melanoma but has limited efficacy against metastatic uveal melanoma, a rare variant arising from the immune privileged eye. To better understand this resistance, we comprehensively profile 100 human uveal melanoma metastases using clinicogenomics, transcriptomics, and tumor infiltrating lymphocyte potency assessment. We find that over half of these metastases harbor tumor infiltrating lymphocytes with potent autologous tumor specificity, despite low mutational burden and resistance to prior immunotherapies.

Methods: Adults with chronic migraine ( = 1,113) were randomized (2:1:1) and treated with double-blind monthly injections of placebo, galcanezumab-120 mg, or galcanenzumab-240 mg for 3 months, followed by a 9-month open-label extension with 120 or 240 mg/month galcanezumab. Headache and medication information was collected by daily eDiary.
Objective: To explore the effect of symptomatic medication overuse (SMO) and headache type on occipital nerve block (ONB) efficacy.
Methods: We conducted a chart review of all of the ONBs performed in our clinic over a 2-year period.
Results: Of 108 ONBs with follow-up data, ONB failed in 22% of injections overall.
Hypomagnesemia has previously been recognized as an uncommon cause of seizures. The electrolyte abnormality is caused by poor gastrointestinal absorption or excessive renal wasting, both from a variety of causes. We report on a 5-week-old patient who developed hypomagnesemic seizures as a consequence of renal magnesium wasting.
