Background: β-lapachone (β-lap) is a naphthoquinone widely found in species of vegetables. However, its poor aqueous solubility limits its systemic administration and clinical applications in vivo. To overcome this limitation, several studies have been carried out in order to investigate techniques that can enhance the solubility and dissolution rate of β-lap, such as the use of inclusion complexes with cyclodextrin.

Article Synopsis
  • Efavirenz is the most commonly used drug for treating AIDS, but its low solubility in water poses challenges for pediatric patients due to limited available formulations.
  • Researchers aimed to improve efavirenz's solubility by creating orodispersible tablets using β-cyclodextrin and polyvinylpyrrolidone, which dissolve quickly in the mouth and enhance drug delivery.
  • Results showed significant improvements in drug solubility, dissolution rates, and overall tablet quality, making this formulation potentially more effective for pediatric patients.

Efavirenz (EFZ) is one of the most used drugs in the treatment of AIDS and is the first antiretroviral choice. However, since it has low solubility, it does not exhibit suitable bioavailability, which interferes with its therapeutic action and is classified as a class II drug according Biopharmaceutical Classification System (low solubility and high permeability). Among several drug delivery systems, the multicomponent systems with cyclodextrins and hydrophilic polymers are a promising alternative for increasing the aqueous solubility of the drug.
