In the 2018 yellow fever (YF) outbreak in Brazil, we generated new transcriptomic data and combined it with clinical and immunological data to decode the pathogenesis of YF. Analyzing 79 patients, we found distinct gene expression patterns between acute YF, other viral infections, and the milder YF-17D vaccine infection. We identified a critical role for low-density, immature neutrophils in severe outcomes, marked by the downregulation of genes essential for neutrophil migration and maturation, such as PADI4, CSF3R, and ICAM1, in deceased patients.

Article Synopsis
  • - Visceral leishmaniasis (VL) is a significant neglected tropical disease where the interaction between immune cells and the parasite is key in determining infection and immune response; type-1 T-cells promote inflammation, while type-2 T-cells support infection establishment.
  • - Recent research highlights the role of mucosal-associated invariant T (MAIT) cells in VL, showing that they can produce essential inflammatory cytokines upon activation, impacting the immune response to the disease.
  • - Findings indicate that VL patients exhibit more activated neutrophils than non-infected individuals, and blocking MR1 can reduce critical inflammatory markers and shift cytokine production towards a detrimental type-2 response, suggesting MR1 involvement in protective immunity against infection.

Zika virus (ZIKV), an arbovirus from the Flaviviridae family, is the causative agent of Zika fever, a mild and frequent oligosymptomatic disease in humans. Nonetheless, on rare occasions, ZIKV infection can be associated with Guillain-Barré Syndrome (GBS), and severe congenital complications, such as microcephaly. The oligosymptomatic disease, however, presents symptoms that are quite similar to those observed in infections caused by other frequent co-circulating arboviruses, including dengue virus (DENV).

Introduction And Methods: In this present work, coronavirus subfamilies and SARS-CoV-2 Variants of Concern (VOCs) were investigated for the presence of MHC-I immunodominant viral peptides using in silico and tools.

Results: In our results, HLA-A*02 haplotype showed the highest number of immunodominant epitopes but with the lowest combined prediction score. Furthermore, a decrease in combined prediction score was observed for HLA-A*02-restricted epitopes when the original strain was compared to the VOCs, indicating that the mutations on the VOCs are promoting escape from HLA-A2-mediated antigen presentation, which characterizes a immune evasion process.
