Monoclonal antibody (mAb) drugs are clinically important for the treatment of various diseases. TTAC-0001 is under development as a new anti-cancer antibody drug targeting VEGFR-2. As the less severe toxicity of TTAC-0001 compared to Bevacizumab, likely due to the decreased in vivo half-life, seems to be related to its structural flexibility, it is important to map the exact flexible regions.

The 3D8 single chain variable fragment (3D8 scFv) is an anti-nucleic acid antibody that can hydrolyze nucleic acids and enter the cytosol of cells without reaching the nucleus. The Tat peptide, derived from the basic region of the HIV-1 Tat protein, translocates to cell nuclei and has TAR RNA binding activity. In this study, we generated a Tat-grafted antibody ((H₃)Tat-3D8) by replacing complementarity-determining region 3 (CDR3) within the VH domain of the 3D8 scFv with a Tat₄₈₋₆₀ peptide (GRKKRRQRRRPPQ).

In this study, the anabolic activities of 89 natural compounds were evaluated by measuring the amount of newly synthesized calcium in the differentiation process of mouse osteoblastic MC3T3-E1 subclone 4 cells. Of these compounds, a low concentration (up to 5 microm) of 3-carene, which is a bicyclic monoterpene in essential oils extracted from pine trees, was shown to stimulate significantly the activity and expression of alkaline phosphatase, an early phase marker of osteoblastic differentiation, on differentiation day 9.
