Introduction: Cabotegravir, an integrase strand transfer inhibitor, and rilpivirine, an NNRTI, constitute the first long-acting (LA), injectable, two-drug ART regimen approved for the maintenance of virological suppression in persons living with HIV-1 (PLHIV). The aim of this study was to assess clinical effectiveness and tolerability of LA cabotegravir/rilpivirine in a real-world setting.

Patients And Methods: We conducted a retrospective, single centre study, including all PLHIV receiving LA cabotegravir/rilpivirine as standard-of-care in our tertiary centre even if initiated in clinical trials.

Article Synopsis
  • The study focuses on endocarditis caused by a specific micro-organism, highlighting its high relapse rate and serious implications for patient management.
  • A retrospective analysis was conducted on 54 hospitalized patients, mostly men aged around 75, revealing that a significant portion had previous histories of endocarditis or heart surgery, with the aortic valve being the most affected.
  • Results indicate that although many patients required surgery, only half underwent the procedure, and 29% needed suppressive antibiotic therapy, with a 6% relapse rate observed over the study period.

Article Synopsis
  • APOBEC3 proteins create some damaged HIV viruses in people who are infected, and this study looked at how these changed when patients switched treatments to dolutegravir and lamivudine.
  • Researchers collected blood samples from patients before and after starting the new treatment and found that the number of defective viruses wasn't drastically lower after a year, but more patients had certain mutations related to drug resistance.
  • Overall, they noticed changes in the mutations, but more research is needed to see what happens in the long run.

The replication-dependent histone H3.1 variant, ubiquitous in multicellular eukaryotes, has been proposed to play key roles during chromatin replication due to its unique expression pattern restricted to the S phase of the cell cycle. Here, we describe recent discoveries in plants regarding molecular mechanisms and cellular pathways involving H3.

Objectives: To minimize confounding factors, we aimed to describe the changes in weight and body mass index (BMI) following the single substitution of tenofovir disoproxil fumarate (TDF) by tenofovir alafenamide (TAF) in people living with HIV (PLWH).

Methods: We designed a retrospective study in a large French cohort. We included all HIV-suppressed adults under TDF + emtricitabine + rilpivirine or elvitegravir/cobistat, who experienced a first switch from TDF to TAF, while other antiretrovirals remained unchanged (Switch group).

Background: Fetal alcohol spectrum disorder (FASD) resulting from prenatal alcohol exposure (PAE) is a common neurodevelopmental disorder, but substantial interindividual heterogeneity complicates timely and accurate assessment, diagnosis, and intervention. The current study aimed to identify classes of children and adolescents with PAE assessed for FASD according to their pattern of significant neurodevelopmental functioning across 10 domains using latent class analysis (LCA), and to characterize these subgroups across clinical features.

Methods: Data from the Canadian National FASD Database, a large ongoing repository of anonymized clinical data received from diagnostic clinics across Canada, was analyzed using a retrospective cross-sectional cohort design.

Objectives: Monkeypox, a zoonotic orthopoxvirus, has spread to many countries in recent months, involving mostly men who have sex with men with multiple partners. Clinical presentation includes skin lesions, systemic signs, and less frequent skin superinfections or anorectal and ophthalmic involvements. We aim to detail cases of myocarditis attributable to monkeypox, an entity that has been poorly described.

Background: The rationale behind the use of ethambutol in the standard tuberculosis treatment is to prevent the emergence of resistance to rifampicin in case of primary resistance to isoniazid. We evaluated whether early detection of isoniazid resistance using molecular testing allows the use an ethambutol-free regimen.

Methods: FAST-TB, a phase 4, French, multicenter, open-label, non-inferiority trial, compared 2 strategies: (1) polymerase chain reaction (PCR)-based detection of isoniazid and rifampicin resistance at baseline using Genotype MTBDR version 2.

Article Synopsis
  • A 10-year retrospective study in France compared the effectiveness and safety of raltegravir and dolutegravir against efavirenz in patients with both HIV-1 and tuberculosis (TB) who started treatment with a rifampicin-based regimen.
  • The study included 117 patients, and after 48 weeks, virological success rates were similar: 61.5% for raltegravir, 63.2% for dolutegravir, and 69.5% for efavirenz, indicating no significant differences (P = 0.68).
  • Additionally, the rates of treatment discontinuation due to adverse events were comparable across the three medications, suggesting similar safety profiles.

The electronic and magnetic structures of diamond nanoparticles with a hydrogenated surface are investigated as a function of annealing temperature under vacuum annealing up to 800-1000 °C. Near edge X-ray absorption fine structure (NEXAFS) spectra together with elemental analysis show successive creation of defect-induced nonbonding surface states at the expense of surface-hydrogen atoms as the annealing temperature is increased above 800 °C. Magnetization and ESR spectra confirm the increase in the concentration of localized spins assigned to the nonbonding surface states upon the increase of the annealing temperature.
