Context And Objectives: The human calcium-sensing receptor (CaSR) plays a key role in calcium homeostasis, and most identified CASR variants are associated with hypercalcemic and hypocalcemic disorders. Here we characterized the pharmacological implications of five heterozygous CASR variants from individuals with familial hypocalciuric hypercalcemia 1 [FHH1: Y63C, I81T, Q459R, W818stop] or autosomal dominant hypocalcemia 1 [ADH1: R955stop].

Methods: Total and cell surface expression levels of wild-type (WT) and variant CaSRs expressed in human embryonic kidney 293T (HEK293T) cells were determined using ELISA, and the pharmacological properties of the receptors were delineated in two functional assays.

Lifestyle factors in the biomedical literature: an ontology and comprehensive resources for named entity recognition.

Motivation: Despite lifestyle factors (LSFs) being increasingly acknowledged in shaping individual health trajectories, particularly in chronic diseases, they have still not been systematically described in the biomedical literature. This is in part because no named entity recognition (NER) system exists, which can comprehensively detect all types of LSFs in text. The task is challenging due to their inherent diversity, lack of a comprehensive LSF classification for dictionary-based NER, and lack of a corpus for deep learning-based NER.

Motivation: Despite significant progress in biomedical information extraction, there is a lack of resources for Named Entity Recognition (NER) and Named Entity Normalization (NEN) of protein-containing complexes. Current resources inadequately address the recognition of protein-containing complex names across different organisms, underscoring the crucial need for a dedicated corpus.

Results: We introduce the Complex Named Entity Corpus (CoNECo), an annotated corpus for NER and NEN of complexes.

Behavioral Coding of Captive African Elephants (): Utilizing DeepLabCut and Create ML for Nocturnal Activity Tracking.

This study investigates the possibility of using machine learning models created in DeepLabCut and Create ML to automate aspects of behavioral coding and aid in behavioral analysis. Two models with different capabilities and complexities were constructed and compared to a manually observed control period. The accuracy of the models was assessed by comparison with manually scoring, before being applied to seven nights of footage of the nocturnal behavior of two African elephants ().

Stimulation of Seed Germination at Suboptimal Temperatures through Biopriming with Biofilm-Forming PGPR KT2440.

This study investigated the germination response to temperature of seeds of nine ecotypes. They are characterized by a similar temperature dependency of seed germination, and 10 °C and 29 °C were found to be suboptimal low and high temperatures for all nine ecotypes, even though they originated from regions with diverse climates. We tested the potential of four PGPR strains from the genera and to stimulate seed germination in the two ecotypes under these suboptimal conditions.

The chemokine receptors CCR1 and CCR5 display overlapping expression patterns and ligand dependency. Here we find that ligand activation of CCR5, not CCR1, is dependent on N-terminal receptor O-glycosylation. Release from O-glycosylation dependency is obtained by increasing CCR5 N-terminus acidity to the level of CCR1.
