The lack of an effective printable ink preparation method and the usual mechanically weak performance obstruct the functional 3D printing hydrogel exploitation and application. Herein, we propose a gentle pre-cross-linking strategy to enable a loosely cross-linked cellulose network for simultaneously achieving favorable printability and a strong hydrogel network via mediating the cellulose self-assembly. A small amount of epichlorohydrin is applied to (i) slightly pre-cross-link the cellulose chains for forming the percolating network to regulate the rheological properties and (ii) form the loosely cross-linked points to mediate the cellulose chains' self-assembly for achieving superior mechanical properties.

Intensively studied 3D printing technology is frequently hindered by the effective printable ink preparation method. Herein, we propose an elegant and gentle solvent consumption strategy to slowly disrupt the thermodynamic stability of the biopolymer (polysaccharide: cellulose, chitin, and chitosan) solution to slightly induce the molecule chains to self-assemble into nanostructures for regulating the rheological properties, eventually achieving the acceptable printability. Spin labels based on cinobufagin, a specific inhibitor of the Na,K-ATPase, have proved valuable tools to characterize the binding site of cardiotonic steroids (CTSs), which also constitutes the extracellular cation pathway. Because existing literature suggests variations in the physiological responses caused by binding of different CTSs, we extended the original set of spin-labeled inhibitors to the more potent bufalin derivatives. Positioning of the spin labels within the Na,K-ATPase site was defined and visualized by molecular docking.

The information obtained from crystallized complexes of the Na ,K -ATPase with cardiotonic steroids (CTS) is not sufficient to explain differences in the inhibitory properties of CTS such as stereoselectivity of CTS binding or effect of glycosylation on the preference to enzyme isoforms. The uncertainty is related to the spatial organization of the hydrophilic cavity at the entrance of the CTS-binding site. Therefore, there is a need to supplement the crystallographic description with data obtained in aqueous solution, where molecules have significant degree of flexibility.

Hydrogels combined with complex 3D shapes and robust mechanical properties are extremely desired soft platforms in the fields of biomaterials, recently, 4D printing has been developed to be further shaped to form required patterns. On the basis of the excellent thixotropy of Laponite and the thermal-reversible sol-gel transition of agarose and easy formation of nanofibers below 35 °C, a 4D printing hydrogel (4D Gel) was fabricated by in situ polymerizing acrylamide in the agarose matrix containing Laponite. The experimental results demonstrated that Laponite played an important role in the improvement of 4D printing, such as endowing the ink with shear-thinning behavior to extrude easily and excellent shape stability after printing.
