A sensitive and specific method using liquid chromatography-electrospray tandem mass spectrometry (LC-MS/MS) for the determination of pinaverium bromide in human plasma was developed and validated. Pinaverium bromide and an internal standard (paclitaxel) were isolated from plasma samples by precipitating plasma, and determined by LC-MS/MS in multiple-reaction monitoring mode. The main metabolite of pinaverium bromide and endogenous substances in plasma did not show any interference.

Article Synopsis
  • - The study aimed to measure three neurosteroids—dehydroepiandrosterone (DHEA), pregnenolone (PREG), and allopregnenolone (AP)—in different brain regions of rats using a specialized extraction and analysis method.
  • - A two-step isolation process involving ethyl acetate-n-hexane extraction and solid-phase extraction (SPE) was employed, followed by derivatization and analysis using HPLC-MS, achieving accurate measurements within specific concentration ranges for each steroid.
  • - The results indicated varying concentrations of DHEA, PREG, and AP across rat brain regions, demonstrating the method's effectiveness for simultaneous measurement of these unconjugated steroids.
