To investigate the flavour evolution mechanism of raw Pu-erh tea (RPT) during storage, the volatile and non-volatile compounds of RPT with different storage years (1-10 years) from the same raw material origin, manufacturer, and storage location in Wenshan Prefecture, Yunnan Province, were systematically analysed by HPLC, HS-SPME-GC-MS, and OAV. The results showed that both cluster analyses based on non-volatile and volatile compounds could classify RPT of different storage years into three ageing cycles, with key turning points in the third and eighth years of storage, which is also accompanied by the colour changing from green to orange or brown, the aroma changing from a faint scent to woody and ageing, the astringency diminishing, and the sweet and mellow increasing. Theophylline was identified as the potential marker of RPT stored 1-3 years, while (-)-catechin gallate, (-)-gallocatechin gallate, quercetin, and rutin as those for a storage of 9-10 years.

The improper application of nonsteroidal anti-inflammatory drugs (NSAIDs) presents significant health hazards via vector food contamination. A critical limitation of these traditional existing approaches is their inability to concurrently discern and distinguish among diverse NSAIDs, presenting a notable gap in the analytical capabilities within this domain. Herein, a creative dual-channel fluorescence sensor array was developed for the rapid discrimination and determination of NSAIDs, utilizing complexes of cucurbit[8]uril (CB[8]) with three distinct modified poly(ethylenimines) (PEIs) to address this challenge.

Nanomedicines need to overcome multiple biological barriers in the body to reach the target area. However, traditional nanomedicines with constant physicochemical properties are not sufficient to meet the diverse and sometimes conflicting requirements during in vivo transport, making it difficult to penetrate various biological barriers, resulting in suboptimal drug delivery efficiency. Smart self-transforming nano-systems (SSTNs), capable of altering their own physicochemical properties (including size, charge, hydrophobicity, stiffness, morphology, etc.

Position detection of elements, especially with high-precision and high-efficiency, continue to present challenges in the off-axis three-mirror space optical system to ensure the imaging quality. To test the position of elements in an off-axis three-mirror system, a method based on ResNet50 and Long short-term memory (LSTM) is proposed in this paper. In the proposed method, point spread function (PSF) with different fields of view is extracted as a high dimensional feature vector by the ResNet50 network to achieve high efficiency.

Light serves as an energy source for cell division and expansion during fruit development. Cell expansion significantly influences fruit size and is closely linked to endoreduplication, a unique cell cycle variation characterized by DNA replication without cytokinesis. Paradoxically, under conditions of ample photosynthates, light signaling suppresses cell expansion.

Uniform seed germination is crucial for consistent seedling emergence and efficient seedling production. In this study, we identified a seed-expressed protein in tomato (Solanum lycopersicum), lateral organ boundaries domain 40 (SlLBD40), that regulates germination speed. CRISPR/Cas9-generated SlLBD40 knockout mutants exhibited faster germination due to enhanced seed imbibition, independent of the seed coat.

Article Synopsis
  • The tumor microenvironment (TME) significantly impacts glioma progression, particularly through features like hypoxia and immunosuppression, although the relationship between hypoxia and immune response in glioma is not fully understood.
  • Researchers identified HSPA5 as a key gene linked to glioma prognosis, associated with higher levels in tumors, poor outcomes, and the promotion of malignant traits via the HIF-1α/HSPA5 pathway.
  • Targeting HSPA5 may offer a new therapeutic approach by reducing hypoxia tolerance and modifying tumor-associated macrophages (TAMs) to an M1 phenotype, potentially enhancing anti-tumor immunity.

Genotype imputation is crucial for genome-wide association studies (GWASs), but reference panels and existing benchmarking studies prioritize European individuals. Consequently, it is unclear which publicly available reference panel should be used for Pakistani individuals, and whether ancestry composition or sample size of the panel matters more for imputation accuracy. Our study compared different reference panels to impute genotype data in 1,814 Pakistani individuals, finding the best performance balancing accuracy and coverage with meta-imputation with TOPMed and the expanded 1000 Genomes (ex1KG) reference.

Social competition determines hierarchical social status, which profoundly influences animals' behavior and health. The dorsomedial prefrontal cortex (dmPFC) plays a fundamental role in regulating social competitions, but it was unclear how the dmPFC orchestrates win- and lose-related behaviors through its downstream neural circuits. Here, through whole-brain c-Fos mapping, fiber photometry, and optogenetics- or chemogenetics-based manipulations, we identified anatomically segregated win- and lose-related neural pathways downstream of the dmPFC in mice.
