20-hydroxyecdysone (20E), a natural polyhydroxylated steroid, has exhibited anti-inflammatory effects across various diseases. This study investigates the potential connection between 20E's anti-inflammatory properties and the RNA-binding protein Lin28b, which is notably upregulated in TNF-α-stimulated endothelial cells. Herein, we discovered that 20E can selectively downregulate Lin28b expression without affecting its paralog Lin28a.

Article Synopsis
  • Ubiquitination, a modification affecting protein stability, is impacted by deubiquitinases (DUBs), with PSMD7 being a specific DUB linked to gastric cancer (GC).
  • Studies reveal that PSMD7 is overexpressed in GC tissues and cell lines, correlating with poorer clinical outcomes and survival rates for patients.
  • Knockdown of PSMD7 decreases GC cell proliferation and invasion, while its presence boosts RAD23B levels, enhancing cell survival and resistance to chemotherapy, particularly cisplatin (DDP).

The MECOM gene encoding a zinc finger protein that functions as a transcription factor, was located on chromosome 3q26, and rearrangements of MECOM often cause its overexpression in acute myeloid leukemia (AML). We identified H2AFY as a novel fusion gene partner of MECOM in an elderly male AML patient with cryptic 3q26 rearrangement using the whole transcriptome sequencing, who carried out abnormal karyotype of 46,XY,t(3;5)(q27;q31),add(14)(p11). We validated the existence of the unreported H2AFY-MECOM fusion gene by RT-PCR and Sanger DNA sequencing, and detected mutations of NRAS and BCOR in this patient.
