Neuroblastoma (NB), a common infantile neuroendocrine tumor, presents a substantial therapeutic challenge when MYCN is amplified. Given that the protein structure of N-Myc is disordered, we utilized Alphafold for prediction and GROMACS for optimization of the N-Myc structure, thereby improving the reliability of the predicted structure. The publicly available datasets GSE49710 and GSE73517 were adopted, which contain the transcriptome data of clinical samples from 598 NB patients.

Super-enhancers (SEs) are prevalent in human cancers and are associated with the accumulation of numerous core TFs. SEs drive the expression of core TF genes by delivering robust transcriptional activation signals.

Previous studies have demonstrated the anti-DN efficacy of Schisandra chinensis Fruit Mixture (SM); however, a comprehensive chemical fingerprint is still uncertain, and its mechanism of action, especially the potential therapeutic targets of anti-DN, needs to be further elucidated.
Objective: Potential mechanisms of SM action on DN were explored through network pharmacology and experimental validation.
Background: Acute myeloid leukemia (AML) is a prevalent malignancy of the hematologic system. Despite advancements in therapeutic approaches, significant heterogeneity and therapeutic resistance pose substantial challenges to treatment. Tumors driven by core transcription factors through super-enhancers can establish core transcriptional regulatory circuits (CRCs) that modulate oncogene expression programs.

However, the clinical application of CAP faces challenges, particularly the need for repeated treatments to maintain therapeutic efficacy and the difficulty in controlling the release of reactive oxygen and nitrogen species (ROS/RNS), which can cause tissue damage at high concentrations. In this study, we developed a controlled-release system by integrating CAP with a skin bionics gelatin scaffold (CAP-GS), enabling the sustained, localized release of CAP without the need for repeated applications.

Silicon concentrate obtained by wet high-intensity magnetic separation was used as the raw material, with impurities removed through a two-stage acid leaching process. The effects of acid concentration, temperature, time, and liquid-solid ratio on impurity removal were examined.

Objective: The impact of lipid-lowering medications on chronic kidney disease (CKD) remains a subject of debate. This Mendelian randomization (MR) study aims to elucidate the potential effects of lipid-lowering drug targets on CKD development.
Methods: We extracted 11 genetic variants encoding targets of lipid-lowering drugs from published genome-wide association study (GWAS) summary statistics, encompassing LDLR, HMGCR, PCSK9, NPC1L1, APOB, ABCG5/ABCG8, LPL, APOC3, ANGPTL3, and PPARA.
Background/purpose: Bisphosphonate-related osteonecrosis of the jaw (BRONJ), a complication arising from the use of bisphosphonates (BPs), inflicts long-term suffering on patients. Currently, there is still a lack of effective treatments. This study aimed to explore the preventive effects of propranolol (PRO) on BRONJ in vitro and in vivo, given PRO's potential in bone health enhancement.

Objective: Rosacea is a common chronic inflammatory disorder primarily affecting the face. While inflammatory factors are known to play a pivotal role in its pathogenesis, their causal relationship with rosacea remains unclear. This study employed a two-sample bidirectional Mendelian randomization (MR) analysis to investigate the causal links between systemic inflammatory regulators and rosacea.

Additionally, it explores how the frequency and timing of P-HIPEC influence treatment outcomes.
Methods: A retrospective analysis was conducted on 227 patients with locally AGC who underwent laparoscopic surgery at Maoming People's Hospital from January 2016 to December 2022.
Background: Autogenous tooth transplantation offers significant advantages and promising success rates for replacing non-retainable teeth. This study aimed to investigate the prognostic factors, especially the impact of root canal treatment (RCT), of autotransplanted teeth in an up-to-6-year follow-up cohort of 167 teeth in Southern China.
Methods: We enrolled adult patients from the Southern Medical University-Shenzhen Stomatology Hospital between 2017 and 2023.
Cell cycle dysregulation and the corresponding metabolic reprogramming play significant roles in tumor development and progression. CDK9, a kinase that regulates gene transcription and cell cycle, also induces oncogene transcription and abnormal cell cycle in AML cells. The function of CDK9 for gene regulation in AML cells requires further exploration.

However, its clinical efficacy is often limited by drug resistance and severe systemic side effects, highlighting the urgent need for innovative therapeutic approaches. Despite advancements in cisplatin-based regimens, research on intravesical cisplatin delivery systems remains scarce.

Although recent studies have highlighted the critical role of deubiquitinases in vascular pathophysiology, their specific contribution to KD remains largely unknown. Herein, we investigated the function of the deubiquitinase USP7 in both KD patients and a CAWS-induced KD murine model.

The selective androgen receptor (AR) modulator YK11 promotes the anabolism of muscle cells and osteoblastic precursor cells. However, its effects on bone marrow-derived mesenchymal stem cells (BMSCs) and the repair of cranial bone defects are unknown. Here, the effects of different concentrations of YK11 on the osteogenic differentiation of BMSCs were determined.

Methods: This retrospective study recruited 4074 patients. Clinical diagnostic indicators and clinical diagnostic processes were applied to develop a hierarchical classification method that involved four sub-task classifications.
Clinical studies intensively highlight two critical brain regions, i,e, dorsal lateral prefrontal cortex (DLPFC) (target node) and subgenual anterior cingulate cortex (sgACC) (indirect target node) for the treatment of neuroimaging-guided repetitive transcranial magnetic stimulation (rTMS) in major depressive disorder (MDD). However, it remains unclear whether the clinical rTMS treatment could modulate the activity of the target and indirect target nodes in MDD patients. We aim to identify the rTMS-induced alteration of brain local and functional connectivity (FC) activities in the target and indirect target nodes.
