Objective: Assessing the accuracy of automated EasyNAT system for rapidly detecting paraffin-embedded tissue for the diagnosis of tuberculosis.

Methods: A retrospective analysis was conducted on 134 patients, comprising 101 with confirmed tuberculosis and 33 without tuberculosis, treated at Beijing Chest Hospital, Capital Medical University, between 2018 and 2022.The clinical diagnostic results served as the standard for assessing the diagnostic performance of the EasyNAT system in comparison to quantitative real-time polymerase chain reaction (qPCR) for tuberculosis detection in paraffin-embedded tissues.

Article Synopsis
  • The study investigates the effectiveness of cell-free Mycobacterium tuberculosis DNA (cf-TB) testing as a biomarker for diagnosing tuberculous pleurisy (TP) due to challenges with low bacterial levels in pleural effusion samples.
  • Conducted across 11 research sites in China, the study analyzed PE samples from 1344 patients suspected of TP, using techniques like Xpert MTB/RIF and mycobacterial culture alongside cf-TB testing.
  • Results revealed that cf-TB testing had a significantly higher sensitivity (73.6%) for definite TP compared to Xpert (40.8%) and mycobacterial culture (54.2%), while maintaining 100% specificity across all tests.

Background: Bacteria in lung play an important role in sustaining lung health. Understanding the characteristics of bacteriomes in lesions of pulmonary tuberculosis (TB) patients, who excrete Mycobacterium tuberculosis (MTB), is important for TB prevention and effective treatment.  METHODS: In this study, bacteriomes in lesions from TB patients excreting bacteria (TB-E) and those from TB patients not excreting bacteria (TB-NE) with matched normal lung tissues (NT) were compared by 16S rRNA sequencing.

Next-generation sequencing (NGS) enables rapid identification of common and rare drug-resistant genetic variations from tuberculosis (TB) patients' sputum samples and MTB isolates. However, whether this technology is effective for formalin-fixed and paraffin-embedded (FFPE) tissues remains unclear. An amplicon-based targeted NGS sequencing panel was developed to predict susceptibility to 9 antituberculosis drugs, including 3 first-line drugs, by directly detecting FFPE tissues.
