Linarin (LA), luteolin (LE), chlorogenic acid (CA) and apigenin (AP) are four major flavonoids with various promising bioactivities found in Compositae (COP) species. A reliable, reproducible and accurate method for the simultaneous and quantitative determination of these four major flavonoids by Ultra Performance Liquid Chromatography (UPLC) analysis was developed. Under the target-guidance of HPLC micro-fractionation, two phenolic compounds, three caffeoylquinic acid derivatives and two sesquiterpene were isolated by high-speed countercurrent chromatography (HSCCC) using elution modes of extrusion-elution. A one-step HSCCC isolation method was developed, which included a solvent system of n-hexane-EtOAc-methanol-water at a ratio of 2:8:3:7 (v/v/v/v).
