Objective: Many patients with melanoma treated with immune checkpoint inhibitors (ICIs) do not derive response. Preclinical and retrospective studies identified that inhibition of the cyclooxygenase (COX) pathway may improve response to ICI treatment.

Methods: This prospective single site phase II trial accrued patients with advanced/metastatic melanoma.

Article Synopsis
  • Verrucous carcinoma (VC) is a rare, low-grade type of squamous cell carcinoma that doesn't metastasize, making it different from invasive squamous cell carcinoma (SCC).
  • Distinguishing VC from a benign condition called verrucous hyperplasia (VH) is difficult due to their similar appearance and clinical features.
  • Researchers generated RNA profiling data from patients to identify new diagnostic markers, revealing specific mRNAs and long non-coding RNAs that could help differentiate between VH and VC, potentially improving treatment decisions.

Article Synopsis
  • Immunosuppressive regulatory T cells (Tregs) play a key role in helping tumors evade the immune response, making them a focus for improving cancer treatments.
  • Recent research highlights the diversity and adaptability of Tregs within tumors, indicating their complex function in both immune responses and therapy effectiveness.
  • This review aims to outline important factors that influence Treg differences in the tumor environment and suggests how understanding this can lead to more targeted therapies for cancer immunotherapy.

Context: Hypophysitis is a known immune-related adverse event (irAE) of immune checkpoint inhibitors (CPIs), commonly associated with CTLA-4 inhibitors and less often with PD-1/PD-L1 inhibitors.

Objective: We aimed to determine clinical, imaging, and HLA characteristics of CPI-induced hypophysitis (CPI-hypophysitis).

Methods: We examined the clinical and biochemical characteristics, magnetic resonance imaging (MRI) of the pituitary, and association with HLA type in patients with CPI-hypophysitis.

Cancer immunotherapies can successfully activate immune responses towards certain tumors. However, this can also result in the development of treatment-induced immune-related adverse events (irAEs) in multiple tissues. Growing evidence suggests that cytokine production in response to these therapeutics potentiates the development of irAEs and may have predictive value as biomarkers for irAE occurrence.

Background: Left ventricular (LV) apical rotation and twist can be estimated noninvasively by speckle-tracking echocardiography (STE). In this study, we tested whether apical rotation is an accurate index of LV contractility.

Methods And Results: We measured LV basal and apical rotation by STE in 11 open-chest anesthetized mongrel dogs under 8 different inotropic stages before and after ligation of either left anterior descending (n=6) or circumflex coronary artery (n=5).
