Understanding the evolutionary processes underlying range-wide genomic variation is critical to designing effective conservation and restoration strategies. Evaluating the influence of connectivity, demographic change and environmental adaptation for threatened species can be invaluable to proactive conservation of evolutionary potential. In this study, we assessed genomic variation across the range of Fraxinus latifolia, a foundational riparian tree native to western North America recently exposed to the invasive emerald ash borer (Agrilus planipennis; EAB).

Arteriovenous fistulas of the filum terminale are rare vascular malformations that predominantly affect males and can present with various neurological symptoms. In this study, we indexed previously published cases of filum terminale arteriovenous fistulas demonstrating that endovascular and microsurgical management are both proven to be appropriate and successful treatment modalities with low complication rates. Endovascular treatment is far less invasive; however, it is associated with higher failure rates, which need to be managed surgically.

Purpose: Immune Checkpoints Inhibitors (ICI) have changed the therapeutic landscape of metastatic renal cell carcinoma first-line treatment with complete response (CR) at metastatic sites observed in 10 to 15% of cases. Delayed nephrectomy could be discussed for patients having a clinical benefit from immunotherapy-based treatment. However, it is unclear whether prior immunotherapy exposure adversely influences the complexity of surgery.

Mechanical stresses, including compression, arise during cancer progression. In solid cancer, especially breast and pancreatic cancers, the rapid tumor growth and the environment remodeling explain their high intensity of compressive forces. However, the sensitivity of compressed cells to targeted therapies remains poorly known.

Background: Antiplatelet drugs, such as clopidogrel, ticagrelor, prasugrel, and acetylsalicylic acid, may be associated with a risk of adverse events (AEs). Vanessa's Law was enacted to strengthen regulations to protect Canadians from drug-related side effects (with mandatory reporting of serious adverse events [SAEs]).

Objective: To determine whether Vanessa's Law has led to an increase in SAE reporting among antiplatelet users.

Background And Objectives: The pharmacokinetics (PK) of piperacillin/tazobactam (PIP/TAZ) is highly variable across different patient populations and there are controversies regarding non-linear elimination as well as the fraction unbound of PIP (f). This has led to a plethora of subgroup-specific models, increasing the risk of misusing published models when optimising dosing regimens. In this study, we aimed to develop a single model to simultaneously describe the PK of PIP/TAZ in diverse patient populations and evaluate the current dosing recommendations by predicting the PK/pharmacodynamics (PD) target attainment throughout life.

Glycolysis and mitochondrial oxidative phosphorylation are the two major metabolic pathways for cellular ATP production. The metabolic plasticity displayed by cancer cells allows them to effectively shift between each of these pathways as a means of adapting to various growth conditions, thus ensuring their survival, proliferation and disease progression. Metabolic plasticity also provides cancer cells with the ability to circumvent many traditional monotherapies aimed at only one or the other of the major ATP-producing pathways.

Article Synopsis
  • Hereditary myopathies involve several hundred genetic variants, with Limb-girdle muscular dystrophies (LGMDs) being a diverse group of disorders linked to more than 30 genes, characterized primarily by limb weakness.
  • The study analyzed 2,372 patients across 21 countries to assess the prevalence of LGMD and Pompe disease through next-generation sequencing (NGS), finding that 11% had pathogenic genetic variants, with a high diagnostic effectiveness for LGMD (86.2%).
  • The findings emphasize the importance of including specific genes in NGS panels for diagnosing LGMW, contributing to a better understanding of LGMD and aiding in the identification of late-onset Pompe disease.

Background & Aims: There is still some uncertainty towards the best method to diagnose the sarcopenic obesity, and specifically, altered body composition. A recent international consensus proposes the association of increased fat mass (FM) with reduced skeletal muscle mass adjusted by weight (SMM/W) (method 1). The ratio of fat mass index (FMI)/fat free mass index (FFMI) (method 2) has also been proposed.

The time-lag method is the standard approach for evaluating membrane permeability, diffusivity and solubility in a single gas permeation experiment. The conventional time-lag method relies on accurately monitoring the pressure rise in a constant volume downstream from the membrane following a change in pressure upstream from the membrane. The same information could be extracted from the upstream pressure decay in the same time-lag experiment.

Tool use and language are highly refined human abilities which may show neural commonalities due to their potential reciprocal interaction during evolution. Recent work provided evidence for shared neural resources between tool use and syntax. However, whether activity within the tool-use network also contributes to semantic neural representations of tool nouns remains untested.

Article Synopsis
  • Diabetic ketoacidosis (DKA) is a serious condition that can occur in children with type 1 diabetes, and stopping intravenous insulin can lead to high blood sugar again.
  • This study evaluated two treatment protocols in children recovering from DKA: one where insulin was given after stopping IV infusion, and another where long-acting insulin was given before stopping the IV.
  • Results showed that the new protocol with early long-acting insulin significantly reduced the risk of ketosis rebound compared to the old protocol.

Mitosis is largely controlled by the reversible phosphorylation of effector proteins. The addition or removal of phosphate groups alters the activities of these proteins, resulting in changes in chromosome structure, cytoskeletal dynamics, nuclear envelope integrity, and other transformations that must occur as a cell progresses through mitosis. Drosophila has been instrumental in the elucidation of the molecular mechanisms of mitosis, which are mostly conserved among animals.

Background: An adverse event (AE) is any undesirable medical manifestation in an individual who has received pharmacological treatment. To be considered serious (SAE), it needs to meet minimally one of the severity criteria by Health Canada. The most recent data (2006) suggested that AEs were underreported (<6%) to health authorities.
