Article Synopsis
  • Existing methods for detecting polyhydroxyalkanoates (PHA) in halophiles are often inaccurate or necessitate prior PHA production.
  • A new technique involves amplifying a conserved region of the phaC gene in halophiles, tested on various strains from Indian salt pans, revealing 29 strains as positive for this gene.
  • The new method demonstrated higher accuracy than traditional Nile Red staining, confirming that only the phaC-positive strains actively produced PHA in nutrient-deficient conditions.

Heterotrophic carbon utilizing microbes were acclimatized in the laboratory by inoculating sludge collected from the waste discharge pond of a small-scale rural abattoir in India in a nutrient solution intermittently fed with glucose and ammonium chloride. Cultures of 10 well-developed isolates were selected and grown in a basal medium containing glucose and ammonium chloride. Culture supernatants were periodically analyzed for ammonium nitrogen (NH(4)(+)-N) and chemical oxygen demand (COD).
