Background: Immune checkpoint inhibitors (ICIs) are recommended to treat patients with deficient mismatch repair/microsatellite instability high (dMMR/MSI-H) metastatic colorectal cancer (mCRC). Pivotal trials have fixed a maximum ICI duration of 2 years, without a compelling rationale. A shorter treatment duration has the potential to improve patients' quality of life and reduce both toxicity and cost without compromising efficacy.

Myeloid cell leukemia 1 (MCL-1) is a member of the B-cell lymphoma 2 protein family and has anti-apoptotic functions. Deregulation of MCL-1 has been reported in several cancers, including lung and breast cancer. In the present study, the association of MCL-1 expression with molecular features in colorectal cancer (CRC) has been highlighted.

Article Synopsis
  • Over two decades, initiatives have aimed to enhance STEM undergraduate outcomes, with the inclusive Research Education Community (iREC) emerging as a scalable reform model that supports STEM faculty in implementing course-based research to improve student learning.
  • This study utilized pathway modeling to describe the HHMI Science Education Alliance (SEA) iREC, identifying how faculty engagement leads to sustainable adoption and improvement of new teaching strategies through feedback from over 100 participating faculty members.
  • The findings indicate that iREC fosters a collaborative environment where STEM faculty can share expertise and data, thereby enhancing their teaching practices and contributing to the overall evolution of undergraduate science education.

Background: In patients with colorectal cancer (CRC), the therapeutic effects of conventional immune checkpoint inhibitors targeting the adaptive immune system are largely limited to those with microsatellite instability-high tumors. Meanwhile, new immunotherapies targeting the innate immune system are attracting increasing attention. CD47 is a representative innate immune checkpoint involved in the evasion of tumor cell phagocytosis by macrophages.

During the COVID-19 pandemic, our findings highlight changes in AML management strategies in India. There was a decrease in overall patient registrations, particularly at large referral centers, while smaller centers saw an increase, reflecting a shift towards more localized care. This shift was accompanied by a rise in the use of hypomethylating agents (HMAs).

  • This study evaluated the ecological health of waters in the Saudi Arabian Exclusive Economic Zone using benthic biotic indices from 2013 to 2018, covering 67 sampling locations with various depths and salinity.
  • Researchers found variations in macrobenthic density, averaging 566 individuals per square meter, and measured biodiversity with a Shannon diversity index averaging 4.70.
  • Most locations were deemed slightly disturbed or undisturbed, with 95.5% of sites showing good or high ecological status according to the M-AMBI analysis.

Perumal Kalaiyarasi JayachandranAnaplastic large cell lymphoma (ALCL) is the second most common type of peripheral T cell lymphoma and an aggressive mature T cell lymphoma. About 50 to 70% of systemic ALCLs are anaplastic lymphoma kinase positive (ALK +), the proportion even higher in the pediatric population. The 5-year survival after chemotherapy is around 70 to 80%.

Background: POLE and POLD1 proofreading deficiency (POLE/D1pd) define a rare subtype of ultramutated metastatic colorectal cancer (mCRC; over 100 mut/Mb). Disease-specific data about the activity and efficacy of immune checkpoint inhibitors (ICIs) in POLE/D1pd mCRC are lacking and it is unknown whether outcomes may be different from mismatch repair-deficient (dMMR)/microsatellite instability-high (MSI-H) mCRCs treated with ICIs.

Patients And Methods: In this global study, we collected 27 patients with mCRC harboring POLE/D1 mutations leading to proofreading deficiency and treated with anti-programmed cell death-ligand 1 alone +/- anti-cytotoxic T-lymphocyte antigen-4 agents.

Circadian clock genes are emerging targets in many types of cancer, but their mechanistic contributions to tumor progression are still largely unknown. This makes it challenging to stratify patient populations and develop corresponding treatments. In this work, we show that in breast cancer, the disrupted expression of circadian genes has the potential to serve as biomarkers.

Unlabelled: Early-stage Hodgkin's lymphoma (ESHL) is highly curable, usually with a combination of chemotherapy and radiation. Real-world data may show differences in survival and prognostic factors when compared to clinical trials. There is limited published literature on ESHL from India.

Background: CDC37 is a key determinant of client kinase recruitment to the HSP90 chaperoning system. We hypothesized that kinase-specific dependency on CDC37 alters the efficacy of targeted therapies for metastatic colorectal cancer (mCRC).

Material And Methods: Two independent mCRC cohorts were analyzed to compare the survival outcomes between CDC37-high and CDC37-low patients (stratified by the median cutoff values): the CALGB/SWOG 80405 trial (226 and 207 patients receiving first-line bevacizumab- and cetuximab-containing chemotherapies, respectively) and Japanese retrospective (50 refractory patients receiving regorafenib) cohorts.

Article Synopsis
  • The study investigates the effectiveness of immune checkpoint inhibitors (ICIs) in patients with deficient mismatch repair (dMMR) colorectal and endometrial cancers, focusing on how different types of mismatch repair alterations affect patient responses.
  • Results showed that patients with mutS co-loss (MSH2 and MSH6) had significantly longer median overall survival compared to those with mutL co-loss (MLH1 and PMS2) in both colorectal cancer (54.6 vs. 36 months) and endometrial cancer (81.5 vs. 48.2 months).
  • Additionally, the neoantigen load (NAL) was found to be higher in mutS co-loss cases, suggesting it could be a key factor

Background: The C-C motif chemokine receptor 5 (CCR5)/C-C motif chemokine ligand 5 (CCL5) axis plays a major role in colorectal cancer (CRC). We aimed to characterize the molecular features associated with / expression in CRC and to determine whether / levels could impact treatment outcomes.

Methods: 7604 CRCs tested with NextGen Sequencing on DNA and RNA were analyzed.

Even though, the blue carbon ecosystems are gaining keen research interest around the globe, the carbon stock of South-West coast of India was poorly reported, and this study is a pioneer attempt and will be an important document for filling the gaps in uncertainties in global carbon stock assessment and also will increase knowledge on biomass variability among mangrove species and mangrove habitats. The study also highlighted the vital role of biomass carbon pool for long-term soil carbon burial. We estimated the above and below-ground biomass carbon stock of 13 mangrove species using two common allometric equations and species-specific equations and reported a very high mean total living biomass carbon stock of Kochi mangroves at 237.

In order to better understand the distribution pattern, pollution degree and the submarine groundwater discharge (SGD) of dissolved heavy metals, 15 subterranean estuaries (STEs) along southwest Indian coast were sampled over three contrasting seasons. The average concentration of metals were ranked as, pre-monsoon > monsoon > post-monsoon with 3 to 12-fold higher groundwater metal concentrations than the adjacent seawater. Average SGD derived essential metal fluxes were five times higher than the toxic metal fluxes of which Fe and Zn together contributed >90 %.

We introduce a family of entanglement witnesses for continuous variable systems, which rely on the sole assumption that their dynamics is that of coupled harmonic oscillators at the time of the test. Entanglement is inferred from the Tsirelson nonclassicality test on one of the normal modes, without any knowledge about the state of the other mode. In each round, the protocol requires measuring only the sign of one coordinate (e.
