Genome search for susceptibility loci of common idiopathic generalised epilepsies.

Genetic factors play a major role in the aetiology of idiopathic generalised epilepsies (IGEs). The present genome scan was designed to identify susceptibility loci that predispose to a spectrum of common IGE syndromes. Our collaborative study included 130 IGE-multiplex families ascertained through a proband with either an idiopathic absence epilepsy or juvenile myoclonic epilepsy, and one or more siblings affected by an IGE trait.

[How to ablate: atrial tachycardia].

The ectopic atrial tachycardia is diagnosed in approx. 10 to 15% of supraventricular tachycardias. Since medical therapy is often shown to be ineffective, ablative procedures became especially important in this disease.

Investigation of the kinetics of degradation of hexopyranosylated cytosine nucleosides using liquid chromatography.

Liquid chromatography was used to follow the degradation of hexopyranosylated cytosine nucleosides in buffers of acid, neutral and alkaline pH and of constant ionic strength. The compounds were found to degrade by hydrolysis to cytosine and/or by deamination to the corresponding uracil nucleosides. Degradation in acid is influenced by the number of sugar hydroxyl groups, presence of sugar double bonds and the type of anomer.

Exclusion of deep venous thrombosis with D-dimer testing--comparison of 13 D-dimer methods in 99 outpatients suspected of deep venous thrombosis using venography as reference standard.

In a direct assay comparison we evaluated the diagnostic performance of 10 novel D-Dimer assays for the exclusion of deep venous thrombosis (DVT). In addition, 3 conventional ELISA D-Dimer assays were included as reference tests. The study was performed in 99 consecutive outpatients referred to the emergency department for clinical suspicion of DVT.

Chronic hypoxia stimulates periarterial sympathetic nerve development in chicken embryo.

Background: Epidemiological findings suggest an association between low-for-age birth weight and the risk to develop coronary heart diseases in adulthood. During pregnancy, an imbalance between fetal demands and supply may result in permanent alterations of neuroendocrine development in the fetus. We evaluated whether chronic prenatal hypoxia increases arterial sympathetic innervation.

A Pathotype System to Describe Intraspecific Variation in Pathogenicity of Meloidogyne chitwoodi.

Tests of eight Dutch Meloidogyne chitwoodi isolates to the differential set for host races 1 and 2 in M. chitwoodi provided no evidence for the existence of host race 2 in the Netherlands. The data showed deviations from expected reactions on the differential hosts, which raised doubts of the usefulness of the host race classification in M.

A common mutation (epsilon1267delG) in congenital myasthenic patients of Gypsy ethnic origin.

Objective: Mutation analysis of the acetylcholine receptor (AChR) epsilon subunit gene in patients with sporadic or autosomal recessive congenital myasthenic syndromes (CMS).

Background: The nicotinic AChR of skeletal muscle is a neurotransmitter-gated ion channel that mediates synaptic transmission at the vertebrate neuromuscular junction. Mutations in its gene may cause congenital myasthenic syndromes.

The diabetes-associated 3243 mutation in the mitochondrial tRNA(Leu(UUR)) gene causes severe mitochondrial dysfunction without a strong decrease in protein synthesis rate.

Cells harboring patient-derived mitochondria with an A-to-G transition at nucleotide position 3243 of their mitochondrial DNA display severe loss of respiration when compared with cells containing the wild-type adenine but otherwise identical mitochondrial DNA sequence. The amount and degree of leucylation of tRNA(Leu(UUR)) were both found to be highly reduced in mutant cells. Despite the low level of leucyl-tRNA(Leu(UUR)), the rate of mitochondrial translation was not seriously affected by this mutation.

A downstream CA repeat sequence increases translation from leadered and unleadered mRNA in Escherichia coli.

When placed downstream of the start codon, multimers of the dinucleotide CA stimulated translation from lacZ, gusA and neo mRNAs in the presence or absence of an untranslated leader sequence. Enhanced expression in the absence of a leader and Shine-Dalgarno sequence indicated that stimulation by CA multimers was independent of translation signals contained within the untranslated leader. Multimers of CA stimulated a significantly higher level of lacZ expression than multimers of individual C or A nucleotides.

Complete remission of a diffuse pontine glioma.

A patient is described in whom a large diffuse glioma of the pons extending into the midbrain was diagnosed at the age of 2 years. Biopsy showed a fibrillary astrocytoma. After shunting of a hydrocephalus, the clinical symptoms abated without conventional therapy.

Cerebellar mutism syndrome.

Since 1980, a growing number of pediatric patients with mutism following posterior fossa surgery have been recognized. This syndrome typically affects children and in rare cases young adults who become mute one or two days after tumor operation but do not show disturbances of consciousness or language comprehension. The disorder persists for 1 to 4 months.

Paraneoplastic limbic encephalitis in two teenage girls.

Paraneoplastic neurological disorders represent remote effects of cancer without invasion of tumor cells into the nervous system. Limbic encephalitis is a distinct entity mostly associated with small-cell carcinoma of the lung. We present the cases of two teenage girls who were admitted with clinical symptoms typical for limbic encephalitis.

Combined therapy of medulloblastoma: review of 46 patients treated in a single institution.

Medulloblastoma is the most frequent malignant brain tumor in pediatric patients. Early treatment strategies, combining surgery and radiotherapy alone, resulted in survival rates of about 40% only. In the last 15 years, chemotherapy was used more frequently in combination with surgery and radiotherapy.

Impedance rise, only clue to severe insulation damage in a transvenous single-lead AICD-system.

We report a case of severe insulation damage 10 months after implantation of a fourth-generation single-lead implantable cardioverter defibrillator in a 45-year-old patient with dilative cardiomyopathy. The measured impedance was > 2000 Ohm and decreased to < 100 Ohm after slight pressure was applied to the device. All other parameters including intracardiac ECGs and markers were normal.

Primitive neuroectodermal tumors of the cerebral hemispheres in two siblings with TP53 germline mutation.

AWe report on two siblings (brother and sister) who developed cerebral PNETs at the age of 5 years and 6 months, respectively. Both children were treated by operation followed by polychemotherapy. The brother also received cranio-spinal irradiation.

Evidence for the role of follicle stimulating hormone in maintaining the oocyte meiotic arrest in pre-antral follicles.

The study was designed to clarify the physiological role of FSH in maintaining the meiotic arrest in vitro of oocytes derived from intact and from hypophysectomized hamsters. The sensitivity to FSH in maintaining the meiotic arrest of oocytes during incubation of pre-antral follicles of adult golden hamsters was estimated: log dose-response curves were characterized by an ED50 of about 0.2 pg FSH/ml (= 1 microU).

An AUG initiation codon, not codon-anticodon complementarity, is required for the translation of unleadered mRNA in Escherichia coli.

We determined the in vivo translational efficiency of 'unleadered' lacZ compared with a conventionally leadered lacZ with and without a Shine-Dalgarno (SD) sequence in Escherichia coli and found that changing the SD sequence of leadered lacZ from the consensus 5'-AGGA-3' to 5'-UUUU-3' results in a 15-fold reduction in translational efficiency; however, removing the leader altogether results in only a twofold reduction. An increase in translation coincident with the removal of the leader lacking a SD sequence suggests the existence of stronger or novel translational signals within the coding sequence in the absence of the leader. We examined, therefore, a change in the translational signals provided by altering the AUG initiation codon to other naturally occurring initiation codons (GUG, UUG, CUG) in the presence and absence of a leader and find that mRNAs lacking leader sequences are dependent upon an AUG initiation codon, whereas leadered mRNAs are not.

Different effects of anesthetics on spontaneous leukocyte rolling in rat skin.

In immunological reactions, leukocytes need to travel from the intravascular space through the vessel wall into the surrounding tissue. The first step in this process is leukocyte rolling, which has often been studied in anesthetized animals. In this study, we investigated the effect of pentobarbital, Hypnorm and both components of the latter, fentanyl and fluanisone, on this primary leukocyte-endothelial cell interaction.

RNA polymerase of vesicular stomatitis virus specifically associates with translation elongation factor-1 alphabetagamma for its activity.

An RNA-dependent RNA polymerase is packaged within the virions of purified vesicular stomatitis virus, a nonsegmented negative-strand RNA virus, which carries out transcription of the genome RNA into mRNAs both in vitro and in vivo. The RNA polymerase is composed of two virally encoded polypeptides: a large protein L (240 kDa) and a phosphoprotein P (29 kDa). Recently, we obtained biologically active L protein from insect cells following infection by a recombinant baculovirus expressing L gene.

A positive and reversible relationship between adrenergic nerves and alpha-1A adrenoceptors in rat arteries.

We evaluated the relationship between the presence of adrenergic nerves and the presence of alpha-1 adrenoceptors (alpha-1 AR) in the arterial tree of the rat. The thoracic aorta and the carotid, mammary, renal and femoral arteries were isolated from 20-week-old male WKY rats, along with the superior mesenteric artery and small (first order) and resistance-sized (third order) side branches of this vessel. Norepinephrine content ([NE]) and specific binding of 300 pM [3H]prazosin were determined.

Termination of quiescence in crustacea. The role of transfer RNA aminoacylation in the brine shrimp Artemia.

In quiescent embryos of the brine shrimp Artemia, the level of aminoacylation of transfer RNAs is low. During resumption of development the charging level of transfer RNAs increases, concomitant with the activation of protein synthesis. The total level of charging rises dramatically from an average of 4% to 50% within a period of 24 h of development.

Expression of a streptomycete leaderless mRNA encoding chloramphenicol acetyltransferase in Escherichia coli.

The chloramphenicol acetyltransferase (cat) gene from Streptomyces acrimycini encodes a leaderless mRNA. Expression of the cat coding sequence as a leaderless mRNA from a modified lac promoter resulted in chloramphenicol resistance in Escherichia coli. Transcript mapping with nuclease S1 confirmed that the 5' end of the cat message initiated at the A of the AUG translational start codon.

Treatment of relapsed Langerhans cell histiocytosis by cyclosporin A combined with etoposide and prednisone.

Optimal treatment for Langerhans cell histiocytosis (LCH) has not yet been established. High-risk patients with systemic LCH may have a fatal course of the disease despite intensive treatment. New approaches using cyclosporin A (CSA) showed promising results.
