Nanolipoprotein particles (NLPs), a lipid bilayer-based nanoparticle platform, have recently been developed for delivery of a variety of molecules of therapeutic interest, but their potential to deliver Fabs with valencies that exceed those of current multivalent formats has not yet been evaluated. Here we describe the development, optimization, and characterization of Fab-NLP conjugates. NLPs were generated with maleimide reactive lipids for conjugation to a Fab with a C-terminal cysteine.
View Article and Find Full Text PDFA new bimetallic platform comprising a six-coordinate Fe(ONO) unit bound to an (ONO)M (M = Fe, Zn) has been discovered ((ONO)H = bis(3,5-di--butyl-2-phenol)amine). Reaction of Fe(ONO) with either (ONO)Fe(py) or with (ONO)FeCl under reducing conditions led to the formation of the bimetallic complex Fe(ONO), which includes unique five- and six-coordinate iron centers. Similarly, the reaction of Fe(ONO) with the new synthon (ONO˙)Zn(py) led to the formation of the heterobimetallic complex FeZn(ONO), with a six-coordinate iron center and a five-coordinate zinc center.
View Article and Find Full Text PDFA family of iron(ii) carbonyl hydride complexes supported by either a bifunctional PNP ligand containing a secondary amine, or a PNP ligand with a tertiary amine that prevents metal-ligand cooperativity, were found to promote the catalytic hydrogenation of CO to formate in the presence of Brønsted base. In both cases a remarkable enhancement in catalytic activity was observed upon the addition of Lewis acid (LA) co-catalysts. For the secondary amine supported system, turnover numbers of approximately 9000 for formate production were achieved, while for catalysts supported by the tertiary amine ligand, nearly 60 000 turnovers were observed; the highest activity reported for an earth abundant catalyst to date.
