The C3 protein is the central molecule within the complement system and undergoes proteolytic activation to C3b in the presence of pathogens. Pattern-independent activation of C3 also occurs via hydrolysis, resulting in C3(HO), but the structural details of C3 hydrolysis remain elusive. Here we show that the conformation of the C3(HO) analog, C3MA, is indistinguishable from C3b.

Pellino 3 E3 ligase promotes starvation-induced autophagy to prevent hepatic steatosis.

Nutrient deprivation is a major trigger of autophagy, a conserved quality control and recycling process essential for cellular and tissue homeostasis. In a high-content image-based screen of the human ubiquitome, we here identify the E3 ligase Pellino 3 (PELI3) as a crucial regulator of starvation-induced autophagy. Mechanistically, PELI3 localizes to autophagic membranes, where it interacts with the ATG8 proteins through an LC3-interacting region (LIR).

Adult Attachment Style, Emotion Regulation and Obsessive-Compulsive Disorder-A Preliminary Cross-Sectional Mediational Investigation of an Attachment-Based Model.

There is evidence that emotion regulation plays a role in the aetiology and maintenance of OCD, but knowledge about what impacts emotion dysregulation is limited. Attachment style is related to both emotion regulation and OCD symptoms, but the link between them has not been thoroughly studied. Examining emotion dysregulation within the context of OCD through an attachment theory framework may lead to a better understanding of the aetiology and maintenance of OCD.

Prevalence and incidence of mastocytosis in adults: a Danish nationwide register study.

Mastocytosis is a group of rare heterogeneous diseases with a prevalence previously found to be 10-23 per 100,000 persons. More awareness and improvements in the diagnostic methods in later years have led to more patients being diagnosed. Here, we set out to present the prevalence and incidence rate of mastocytosis among the adult Danish population.

Inpatient Small Bowel Capsule Endoscopy: Not Associated With Bleeding Site Identification or 30-Day Readmission Prevention.

Background The utility of small bowel capsule endoscopy (SBCE) in the inpatient setting is controversial due to retention rates and costs. Aim This study aims to evaluate whether using SBCE significantly improved the identification of potential bleeding sites or reduced the risk of 30-day readmission for overt or occult gastrointestinal bleeding. Methods This was a single-center retrospective cohort study involving inpatients who underwent SBCE at a suburban tertiary care hospital from January 1, 2012, to January 1, 2022, for suspected small bowel bleeding.

A prospective study of male physical activity and fecundability.

Study Question: To what extent is male physical activity (PA) associated with fecundability (per-cycle probability of conception)?

Summary Answer: Preconception levels of vigorous, moderate, or total PA were not consistently associated with fecundability across Danish and North American cohorts, but there was suggestive evidence that bicycling with a 'soft, comfort seat' was associated with reduced fecundability in both cohorts, especially among males with greater BMI.

What Is Known Already: Among males, some studies indicate that moderate PA might improve fertility, whereas vigorous PA, especially bicycling, might be detrimental.

Introduction: Placental-derived extracellular vesicles (EVs) are nano-organelles that facilitate intercellular communication between the feto-placental unit and the mother. We evaluated a novel Multiple Microarray analyzer for identifying surface markers on plasma EVs that predict preterm delivery and preeclampsia compared to term delivery controls.

Material And Methods: In this prospective exploratory cohort study pregnant women between 24 and 40 gestational weeks with preterm delivery (n = 16), preeclampsia (n = 19), and matched term delivery controls (n = 15) were recruited from Bnai Zion Medical Center, Haifa, Israel.

The human gut microbiome is highly personal. However, the contribution of gut physiology and environment to variations in the gut microbiome remains understudied. Here we performed an observational trial using multi-omics to profile microbiome composition and metabolism in 61 healthy adults for 9 consecutive days.

Comparative lifespan and healthspan of nonhuman primate species common to biomedical research.

There is a critical need to generate age- and sex-specific survival curves to characterize chronological aging consistently across nonhuman primates (NHP) used in biomedical research. Sex-specific Kaplan-Meier survival curves were computed in 12 translational aging models: baboon, bonnet macaque, chimpanzee, common marmoset, coppery titi monkey, cotton-top tamarin, cynomolgus macaque, Japanese macaque, pigtail macaque, rhesus macaque, squirrel monkey, and vervet/African green. After employing strict inclusion criteria, primary results are based on 12,269 NHPs that survived to adulthood and died of natural/health-related causes.

The effectiveness of diabetes training of psychiatric health professionals on individuals with diabetes and psychiatric disorders - a pragmatic controlled trial in Denmark.

Background: Individuals with diabetes and co-existing psychiatric disorders have more diabetes complications and lower life expectancy than those with diabetes but no co-existing psychiatric disorders. Psychiatric health professionals may have a role in improving these outcomes but often lack diabetes knowledge and skills. This study aims to examine the effectiveness of a diabetes training course for psychiatric health professionals on their diabetes knowledge and skills and clinical outcomes, diabetes support and diabetes distress among individuals with diabetes and psychiatric disorders treated in psychiatric outpatient clinics.

Temporal trends and sources of organic micropollutants in wastewater.

Effluent wastewater from conventional wastewater treatment plants (WWTPs) is a source of environmental micropollutants. This study investigated temporal trends of organic micropollutants in effluent wastewater, aiming to identify underlying drivers and their implications for treatment efficiency. From September to December 2022, we collected 168 effluent and 10 influent samples.

Impact of laparoscopic ultrasound during PIPAC directed treatment of unresectable peritoneal metastasis.

Objectives: Laparoscopic ultrasound (LUS) combines both laparoscopy and ultrasound imaging of the peritoneum liver and retroperitoneum. LUS has not been described in treatments with pressurized intraperitoneal aerosol chemotherapy (PIPAC). We present our experience with LUS in patients undergoing PIPAC.

Clinical outcome is unlinked to injection of adipose-derived regenerative cells in the axilla of breast cancer-related lymphedema patients.

Background: Injection of autologous adipose-derived regenerative cells (ADRCs) combined with lipotransfer has been suggested to alleviate symptoms in diseases including breast cancer-related lymphedema (BCRL). We recently performed a randomized controlled trial injecting lipoaspirate with ADRCs into the axilla of BCRL patients, and here we aimed in the intervention group to define in an unbiased fashion whether ADRC injection was linked to the clinical outcome.

Methods: 39 BCRL patients received lipotransfer assisted with autologous ADRCs (4.
