In 1995, a committee of the International Headache Society developed and published the first edition of the These have not been revised. With the emergence of new medications, neuromodulation devices and trial designs, an updated version of the International Headache Society is warranted. Given the scarcity of evidence-based data for cluster headache therapies, the update is largely consensus-based, but takes into account lessons learned from recent trials and demands by patients.

Context: Supraphysiological serum insulin-like growth factor-I (IGF-I) concentrations have been a matter of concern in children treated with GH because high IGF-I levels were associated with risk of later disease in former epidemiological studies.

Objective: To determine whether a single IGF-I measurement reliably reflects lifetime IGF-I exposure we evaluated intraindividual longitudinal tracking of IGF-I and IGF-binding protein-3 (IGFBP-3) levels and we estimated cumulative lifetime exposure to IGF-I in healthy and GH-treated individuals.

Methods: We included 6459 healthy participants (cross-sectional = 5326; longitudinal = 1133) aged 0-76 years (9963 serum samples) and 9 patients born small-for-gestational-age (SGA) with 238 serum samples during GH treatment.

Article Synopsis
  • - This study compared traditional instructor-led lessons and immersive virtual reality (IVR) lessons to teach basic point-of-care ultrasound (PoCUS) skills to final-year medical students through a randomized controlled trial.
  • - Learning efficacy was measured using the Objective Structured Assessment of Ultrasound Skills (OSAUS), and results showed no significant differences in performance scores between the two teaching methods.
  • - While both courses had similar maintenance costs, the instructor-led course had much higher startup costs; the findings indicate that IVR could be a viable alternative to traditional instruction, though further research is needed.

Objectives: Opioid use disorder is associated with high rates of mortality and has become an escalating global health issue. Opioid agonist treatment (OAT) with oral methadone or daily sublingual buprenorphine hydrochloride, either administered separately or in combination with naloxone hydrochloride (SL-BPN, SL-BPN/NX), is supervised by a healthcare professional experienced in treating opioid use disorder to ensure proper dosing and prevent misuse. For that reason, there may be substantial direct and indirect costs associated with OAT.

Background And Objective: Within medical research, cure models are useful for analyzing time-to-event data in the scenario where a proportion of the analyzed individuals are expected to never experience the event of interest. Cure models are also useful for modelling the relative survival in scenarios where a proportion of the individuals are expected to eventually experience a mortality rate similar to that of the general population. Here we present two R packages, cuRe and rstpm2, that provide researchers with several tools for performing statistical inference using parametric cure models.

Pathogenic mutations in the BRCA2 tumor suppressor gene predispose to breast, ovarian, pancreatic, prostate, and other cancers. BRCA2 maintains genome stability through homology-directed repair (HDR) of DNA double-strand breaks (DSBs) and replication fork protection. Nonsense or frameshift mutations leading to truncation of the BRCA2 protein are typically considered pathogenic; however, missense mutations resulting in single amino acid substitutions can be challenging to functionally interpret.

Article Synopsis
  • Atypical hemolytic uremic syndrome (aHUS) is a serious condition that can cause organ failure and is linked to issues with the complement system and the CD46 protein.* -
  • A pediatric patient with aHUS was found to have a new variant in the CD46 gene, which led to reduced surface expression and impaired binding to C3b, a key part of the immune response.* -
  • The study identified that the patient had a higher expression of rare CD46 isoforms, which function less effectively, suggesting multiple mechanisms are involved in the dysfunction of CD46 in aHUS.*

Chronic obstructive pulmonary disease (COPD) mortality risk is often estimated using the BODE (body mass index, obstruction, dyspnea, exercise capacity) index, including body mass index, forced expiratory volume in 1 second, dyspnea score, and 6-minute walk distance. Diffusing capacity of the lung for carbon monoxide (Dl) is a potential predictor of mortality that reflects physiology distinct from that in the BODE index. This study evaluated Dl as a predictor of mortality using participants from the COPDGene study.
