Hypoxia-inducible factor prolyl hydroxylase (PHD) inhibitors have been approved for treating renal anemia yet have failed clinical testing for inflammatory bowel disease because of a lack of efficacy. Here we used a multimodel multimodal generative artificial intelligence platform to design an orally gut-restricted selective PHD1 and PHD2 inhibitor that exhibits favorable safety and pharmacokinetic profiles in preclinical studies. ISM012-042 restores intestinal barrier function and alleviates gut inflammation in multiple experimental colitis models.

Article Synopsis
  • Kernel dehydration rate (KDR) significantly influences maize harvesting and kernel quality, but its mechanisms are not fully understood.
  • Researchers discovered a quantitative trait locus (QTL) called qKDR1, which regulates the expression of a peptide gene, RPG, that produces a micropeptide (microRPG1) impacting KDR through ethylene signaling pathways.
  • Knockout experiments showed that loss of microRPG1 leads to quicker KDR, while overexpression or applying the micropeptide slows it down, providing insight for future maize breeding strategies.

Background: An imbalance of antiproliferative BMP (bone morphogenetic protein) signaling and proliferative TGF-β (transforming growth factor-β) signaling is implicated in the development of pulmonary arterial hypertension (PAH). The posttranslational modification (eg, phosphorylation and ubiquitination) of TGF-β family receptors, including BMPR2 (bone morphogenetic protein type 2 receptor)/ALK2 (activin receptor-like kinase-2) and TGF-βR2/R1, and receptor-regulated Smads significantly affects their activity and thus regulates the target cell fate. BRCC3 modifies the activity and stability of its substrate proteins through K63-dependent deubiquitination.

Article Synopsis
  • This study compared the effectiveness of coronary computed tomography angiography (CCTA) and cardiac magnetic resonance (CMR) in diagnosing ischemia in patients with coronary artery disease.
  • Researchers enrolled 92 patients who underwent both CCTA and CMR, along with invasive procedures, to analyze plaque characteristics and blood flow metrics.
  • The findings revealed that CCTA-derived plaque characteristics and CMR-derived myocardial perfusion reserve (MPR) were more accurate in diagnosing significant ischemia than the traditional myocardial blood flow (MBF) measurement.

Human UDP-glucuronosyltransferases (UGTs) play a pivotal role as prominent phase II metabolic enzymes, mediating the glucuronidation of both endobiotics and xenobiotics. Dimerization greatly modulates the enzymatic activities of UGTs. In this study, we examined the influence of three mutations (H35A, H268Y, and N68A/N315A) and four truncations (signal peptide, single transmembrane helix, cytosolic tail, and di-lysine motif) in UGT2B7 on its heterodimerization with wild-type UGT1A9, using a Bac-to-Bac expression system.

Aluminum (alum) adjuvant is the most extensively used protein subunit vaccine adjuvant, and its effectiveness and safety have been widely recognized. The surface charge of the antigen determines its electrostatic adsorption to alum adjuvant, which directly affects the immune efficacy of the protein vaccine. In our study, we precisely modified its surface charge by inserting charged amino acids into the flexible region of the SARS-CoV-2 receptor-binding domain (RBD), achieving electrostatic adsorption and a site-specific anchor between the immunogen and alum adjuvant.

Background Radiofrequency ablation (RFA) is a widely used treatment for atrial fibrillation, reducing the risk of cardiac arrhythmia. Detailed visualization and quantification of atrial scarring has the potential to improve preprocedural decision-making and postprocedural prognosis. Conventional bright-blood late gadolinium enhancement (LGE) MRI can help detect atrial scars; however, its suboptimal myocardium to blood contrast inhibits accurate scar estimation.

The cardiovascular system is hardwired to the brain via multilayered afferent and efferent polysynaptic axonal connections. Two major anatomically and functionally distinct though closely interacting subcircuits within the cardiovascular system have recently been defined: The artery-brain circuit and the heart-brain circuit. However, how the nervous system impacts cardiovascular disease progression remains poorly understood.

Article Synopsis
  • ACE2 is crucial for converting angiotensin II and its stability is influenced by AMPK phosphorylation; however, MDM2 can degrade ACE2 through ubiquitination, potentially linking it to pulmonary hypertension (PH).
  • Bioinformatic analyses and experiments in cultured cells, mouse models, and human samples were utilized to study the interaction between AMPK and MDM2's effects on ACE2 in PH.
  • Results showed that increased MDM2 and decreased ACE2 levels are associated with idiopathic pulmonary arterial hypertension, and modulating ACE2's phosphorylation and ubiquitination could mitigate PH, highlighting their roles in its pathogenesis.
