VISTA is a key immune checkpoint receptor under investigation for cancer immunotherapy; however, its signaling mechanisms remain unclear. Here we identify a conserved four amino acid (NPGF) intracellular motif in VISTA that suppresses cell proliferation by constraining cell-intrinsic growth receptor signaling. The NPGF motif binds to the adapter protein NUMB and recruits Rab11 endosomal recycling machinery.

Exceptionally diverse type V CRISPR-Cas systems provide numerous RNA-guided nucleases as powerful tools for DNA manipulation. Two known Cas12e nucleases, DpbCas12e and PlmCas12e, are both effective in genome editing. However, many differences exist in their in vitro dsDNA cleavage activities, reflecting the diversity in Cas12e's enzymatic properties.

Diamond is an exceptional material with great potential across various fields owing to its interesting properties. However, despite extensive efforts over the past decades, producing large quantities of desired ultrathin diamond membranes for widespread use remains challenging. Here we demonstrate that edge-exposed exfoliation using sticky tape is a simple, scalable and reliable method for producing ultrathin and transferable polycrystalline diamond membranes.

Background: Anterior shoulder instability is a common pathology seen especially in young men and highly active patient populations. Subluxation is a commonly encountered clinical issue, yet little is known about the effects of first-time subluxation compared with dislocation on shoulder stability and clinical outcomes after surgical stabilization.

Purpose: To compare revision and redislocation rates as well as patient-reported outcomes (PROs) between subluxators and dislocators after a first-time anterior shoulder instability event.

Background: Patients undergoing bariatric surgery may be at increased risk for postsurgical opioid dependence, highlighting a need for opioid-sparing anesthesia.

Objectives: Liposomal bupivacaine (LB), a prolonged release formulation of bupivacaine, may improve postoperative pain management and reduce postsurgical opioid use. This retrospective claims-database study investigated the effects of LB versus non-LB analgesia on opioid use and healthcare resource utilization (HCRU) in patients receiving laparoscopic sleeve gastrectomy (SG).

As patients with relapsed/refractory multiple myeloma (RRMM) continue to live longer, they might get exposed to most available drugs and drug classes during the disease course. For such late line RRMM or among patients without access to novel therapies, retreatment with a drug that the disease had previously been refractory to might be one option. In this retrospective study, we describe 315 patients with RRMM at our institution who were retreated with a drug that the disease had been previously refractory to.

Elucidating details of biology's selective uptake and trafficking of rare earth elements, particularly the lanthanides, has the potential to inspire sustainable biomolecular separations of these essential metals for myriad modern technologies. Here, we biochemically and structurally characterize () LanD, a periplasmic protein from a bacterial gene cluster for lanthanide uptake. This protein provides only four ligands at its surface-exposed lanthanide-binding site, allowing for metal-centered protein dimerization that favors the largest lanthanide, La.

  • Coronary computed tomography angiography (CCTA) is used to evaluate cardiovascular risk by quantifying coronary plaque, and deep learning technology helps automate this process.
  • A study involving 2803 patients analyzed how age and sex affect coronary plaque volume and its relation to the risk of myocardial infarction, showing that plaque volume increases with age and is typically higher in men.
  • Patients with coronary plaque in the ≥75th percentile were found to have a significantly higher risk of myocardial infarction compared to those below the 50th percentile, suggesting that deep learning-based plaque measurements can effectively predict cardiac events.

Advanced n-type organic electrochemical transistors (OECTs) play an important part in bioelectronics, facilitating the booming of complementary circuits-based biosensors. This necessitates the utilization of both n-type and p-type organic mixed ionic-electronic conductors (OMIECs) exhibiting a balanced performance. However, the observed subpar electron charge transport ability in most n-type OMIECs presents a significant challenge to the overall functionality of the circuits.

Background: Antibodies against the N-methyl-D-aspartate receptor (NMDAR) have been described in the serum of people with schizophrenia spectrum disorders (schizophrenia). However, the prevalence and clinical relevance of these antibodies in schizophrenia is unclear. This knowledge gap includes the possibility of such antibodies being associated with a distinct clinical profile, which in turn might warrant a distinct treatment approach.

  • Three generations of tyrosine kinase inhibitors (TKIs) exist for ALK fusion-positive non-small cell lung cancer, but they fail to effectively address resistance, brain activity, and TRK inhibition issues.* -
  • NVL-655, a new TKI, shows superior selectivity and potency against ALK mutations, significantly outperforming current approved ALK TKIs in preclinical studies.* -
  • Preliminary results from a phase I/II trial indicate NVL-655's promise for treating heavily pretreated patients, including those with brain metastases and resistance mutations, potentially making it a fourth-generation advancement for ALK-driven cancers.*

  • Patients with non-small cell lung cancer (NSCLC) who have specific mutations (-Mut) are often in a more advanced stage of the disease and have a poorer prognosis; however, there is limited research on this group due to the rarity of the mutations.
  • The study utilized data from The Cancer Genome Atlas (TCGA) and a clinical center to analyze clinical, pathological, and molecular characteristics of NSCLC -Mut patients, examining 481 samples from TCGA and 20 patients from the center.
  • Results indicated that -Mut patients had a significantly worse prognosis compared to wild-type (WT) patients, but those with Napsin A expression experienced longer overall survival, suggesting that Napsin A could be a potential prognostic

Article Synopsis
  • ROS1 gene fusions account for 1%-2% of non-small cell lung cancer (NSCLC) and represent a significant oncogenic driver, changing how this cancer is treated with targeted therapies.
  • First-line FDA-approved treatments like crizotinib, entrectinib, and repotrectinib have improved outcomes for patients, but resistance to these drugs is a common issue that can lead to disease relapse.
  • Ongoing research into resistance mechanisms has led to the development of next-generation ROS1 TKIs and new treatment strategies, prompting a careful selection of therapies to enhance patient care and extend survival for those with metastatic and non-metastatic ROS1+ NSCLC.

Size-dependent phagocytosis is a well-characterized phenomenon in monocytes and macrophages. However, this size effect for preferential gene delivery to these important cell targets has not been fully exploited because commonly adopted stabilization methods for electrostatically complexed nucleic acid nanoparticles, such as PEGylation and charge repulsion, typically arrest the vehicle size below 200 nm. Here, we bridge the technical gap in scalable synthesis of larger submicron gene delivery vehicles by electrostatic self-assembly of charged nanoparticles, facilitated by a polymer structurally designed to modulate internanoparticle Coulombic and van der Waals forces.

Deubiquitinase-targeting chimera (DUBTAC) is a promising technology for inducing targeted protein stabilization (TPS). Despite its therapeutic potential, very few proteins have been stabilized by DUBTACs to date. The limited applicability of this technology is likely due to the modest DUBTAC-induced protein stabilization effect, and the scarcity of effective deubiquitinase ligands that can be harnessed for DUBTAC development.

Article Synopsis
  • - The study evaluated how various cardiometabolic risk factors, such as metabolic syndrome and individual factors like high fasting glucose and low HDL cholesterol, impact the progression of coronary plaque and the likelihood of major cardiovascular events in patients with stable coronary artery disease.
  • - In a cohort of 1,200 patients followed over several years, 28% experienced rapid coronary plaque progression, identified primarily through increased atheroma volume, which indicates worsening arterial health.
  • - The findings suggest that a combination of risk factors, particularly high blood sugar and blood pressure, significantly forecast both plaque progression and serious cardiovascular incidents, highlighting the importance of monitoring these factors in at-risk patients.
