Background: Low-dose chest computed tomography (LDCT) screening improves early detection of lung cancer but poses challenges such as false positives and overdiagnosis, especially for nodules smaller than 8 mm where follow-up guidelines are unclear. Traditional risk prediction models have limitations, and deep learning (DL) algorithms offer potential improvements but often require large datasets. This study aimed to develop a DL-based, label-free lung cancer risk prediction model using alternative LDCT images and validate it in individuals without non-calcified solid pulmonary nodules larger than 8 mm.

Background: The association, if any, between the transmitral mean pressure gradient (TMPG) after mitral transcatheter edge-to-edge repair and 1-year mortality is controversial in patients undergoing mitral transcatheter edge-to-edge repair with the MitraClip system. We sought to estimate the association between intraoperatively measured residual mitral regurgitation (rMR) and TMPG and 1-year mortality among patients undergoing mitral transcatheter edge-to-edge repair to facilitate decisions on additional devices.

Methods: In patients with severe secondary (functional) MR, we analyzed registry data using generalized estimating equations.

Successful pancreatic ductal adenocarcinoma (PDAC) immunotherapy requires therapeutic combinations that induce quality T cells. Tumor microenvironment (TME) analysis following therapeutic interventions can identify response mechanisms, informing design of effective combinations. We provide a reference single-cell dataset from tumor-infiltrating leukocytes (TILs) from a human neoadjuvant clinical trial comparing the granulocyte-macrophage colony-stimulating factor (GM-CSF)-secreting allogeneic PDAC vaccine GVAX alone, in combination with anti-PD1 or with both anti-PD1 and CD137 agonist.

Despite the broad catalytic relevance of metal-support interfaces, controlling their chemical nature, the interfacial contact perimeter (exposed to reactants), and consequently, their contributions to overall catalytic reactivity, remains challenging, as the nanoparticle and support characteristics are interdependent when catalysts are prepared by impregnation. Here, we decoupled both characteristics by using a raspberry-colloid-templating strategy that yields partially embedded PdAu nanoparticles within well-defined SiO or TiO supports, thereby increasing the metal-support interfacial contact compared to nonembedded catalysts that we prepared by attaching the same nanoparticles onto support surfaces. Between nonembedded PdAu/SiO and PdAu/TiO, we identified a support effect resulting in a 1.

Article Synopsis
  • - The study introduces the SPRINTER algorithm, which analyzes single-cell DNA sequencing to identify and classify the proliferation rates of different cancer cell clones within tumors, shedding light on the variability of cell growth among these clones.
  • - Applying SPRINTER to nearly 15,000 non-small cell lung cancer cells showed significant differences in clone proliferation, which was corroborated by various imaging techniques and indicated that more proliferative clones also had a higher likelihood of metastasis and altered genetic replication patterns.
  • - The algorithm's effectiveness was further demonstrated in breast and ovarian cancer datasets, where it uncovered higher proliferation rates and genetic variations in specific, more rapidly growing cell clones.

Background And Purpose: Rescue stent (RS) is an accepted rescue option after failed mechanical thrombectomy (MT) for acute ischemic stroke due to intracranial atherosclerotic stenosis (ICAS)-related large vessel occlusion (LVO). However, the long-term outcomes (≥ 12 months) of RS have not yet been elucidated.

Materials And Methods: We retrospectively analyzed the data of 154 patients with RS for ICAS-related LVO, which were identified from prospectively maintained multicenter database of RS after MT failure, to assess good outcome (mRS 0-2), mortality, stroke recurrence, symptomatic intracranial hemorrhage (SICH) and stent patency.

Article Synopsis
  • - The study focused on analyzing the occurrence and timing of growth in subsolid nodules (SSNs) that had been stable for five years, investigating their clinical significance and determining an effective follow-up strategy.
  • - Out of 235 SSNs followed for up to 181 months after initial stability, only 14 (6.0%) showed growth, and no metastases were found, indicating a very low growth incidence.
  • - The proposed follow-up with CT scans every five years post-initial stability could effectively detect most of the growing nodules and any clinical stage shifts.

Background: Human papilloma virus (HPV)-positive oropharyngeal squamous cell carcinoma (OPSCC) is rising in prevalence and is associated with better survival than HPV-negative OPSCC. In surgically treated HPV-negative OPSCCs, adverse pathologic features such as positive surgical margins, extra-nodal extension (ENE) and perineural invasion are well described to portend worse clinical outcomes. These adverse pathological features, however, are not validated prognostic markers among surgically treated HPV-positive OPSCCs.

Article Synopsis
  • Many non-small-cell lung cancer (NSCLC) patients develop resistance to existing EGFR tyrosine kinase inhibitors (TKIs), particularly due to _C797S mutations that render osimertinib ineffective.
  • BLU-945 is a new orally available EGFR-TKI that effectively targets both activating and resistance mutations, including EGFR_C797S, showing significant potency while sparing healthy EGFR.
  • In preclinical and early clinical studies, BLU-945 demonstrated promising results, inhibiting tumor growth in resistant models and patients, suggesting it could be a viable treatment option for NSCLC patients who no longer respond to osimertinib.

Mesenchymal stromal cells (MSCs) display heterogeneity in origin and functional role in tissue homeostasis. Subsets of MSCs derived from the neural crest express nestin and serve as niches in bone marrow, but the possibility of coaxing MSCs into nestin-expresing cells for enhanced supportive activity is unclear. In this study, as an approach to the chemical coaxing of MSC functions, we screened libraries of clinically approved chemicals to identify compounds capable of inducing nestin expression in MSCs.

Article Synopsis
  • A study was conducted to explore how user experience and characteristics of polyps affect interactions with computer-aided detection (CADe) systems during colonoscopy.
  • The CADe system, built using YOLOv4 and trained on nearly 18,000 images, showed an improvement in polyp detection accuracy from 69.7% to 77.7% when assisted by CADe, although misdetections could decrease accuracy.
  • The results indicated that less experienced users (like nurses) benefited more from CADe assistance compared to experts, and while overall sensitivity improved for easier polyp detection, it was lower for more challenging lesions like sessile serrated lesions (SSLs).

Microglia replacement strategies are increasingly being considered for the treatment of primary microgliopathies like adult-onset leukoencephalopathy with axonal spheroids and pigmented glia (ALSP). However, available mouse models fail to recapitulate the diverse neuropathologies and reduced microglia numbers observed in patients. In this study, we generated a xenotolerant mouse model lacking the fms-intronic regulatory element (FIRE) enhancer within Csf1r, which develops nearly all the hallmark pathologies associated with ALSP.
