Article Synopsis
  • Prognostic biomarkers are important for understanding sporadic desmoid tumors (DT) due to their unpredictable nature; this study focused on gene expression differences between DTs located in the thoracic versus abdominal wall.
  • A retrospective analysis of 197 sporadic DT patients showed significant relationships between relapse-free survival (RFS) and factors like tumor size, location, and specific gene mutations, notably the CTNNB1 T41A and S45F mutations.
  • Results suggest that tumors in the abdominal wall have a better prognosis compared to extra-abdominal locations, with distinct gene expression profiles influencing cancer pathways related to their behavior and outcomes.

Major depressive disorder (MDD) is a heterogeneous clinical syndrome with widespread subtle neuroanatomical correlates. Our objective was to identify the neuroanatomical dimensions that characterize MDD and predict treatment response to selective serotonin reuptake inhibitor (SSRI) antidepressants or placebo. In the COORDINATE-MDD consortium, raw MRI data were shared from international samples ( = 1,384) of medication-free individuals with first-episode and recurrent MDD ( = 685) in a current depressive episode of at least moderate severity, but not treatment-resistant depression, as well as healthy controls ( = 699).

Background: Variants in and (encoding apolipoprotein E and presenilin 1, respectively) alter the risk of Alzheimer's disease. We previously reported a delay of cognitive impairment in a person with autosomal dominant Alzheimer's disease caused by the variant who also had two copies of the apolipoprotein E3 Christchurch variant ( ). Heterozygosity for the variant may influence the age at which the onset of cognitive impairment occurs.

Article Synopsis
  • A study conducted between 1998 and 2020 identified 43 new species in diverse ecosystems across Europe, Asia, and the Americas, using genetic analysis to classify them into existing and new subclades within a major clade.
  • The evolutionary history of this clade includes separations before and after the breakup of Gondwana, leading to distinct groups with varying lifestyles, such as soilborne and aerial species.
  • High levels of hybridization were observed in some subclades, indicating that environmental and breeding systems may encourage the emergence of new species, revealing significant gaps in our understanding of potential pathogen threats to ecosystems.

Article Synopsis
  • - The study examined how complete revascularization after percutaneous coronary intervention (PCI) impacts patients with left main coronary artery disease, using data from the EXCEL trial involving 903 patients over a 5-year follow-up.
  • - Findings showed that the risk of death or myocardial infarction (MI) did not significantly differ based on traditional measures of complete revascularization but was notably higher with a higher residual Jeopardy Score (rJS), particularly for patients with untreated severe stenoses in the left circumflex artery (LCX).
  • - Specifically, patients with an rJS of 4 or more had worse outcomes, with increased mortality and spontaneous MI, highlighting the importance of addressing untreated high-grade lesions post-PCI for

Article Synopsis
  • This study investigates the effects of high ratio fresh frozen plasma (FFP) to red blood cell (RBC) transfusions in children experiencing shock due to trauma, aiming to clarify their outcomes compared to low ratio transfusions.
  • An analysis of data from 135 injured children showed that while more severe injuries were present in the high ratio group, there was no significant difference in mortality rates or extended hospital stays between those receiving high and low ratios of FFP/RBC.
  • The findings indicate that high ratio FFP/RBC transfusion does not lead to worse outcomes, highlighting variability in massive transfusion protocols across different medical institutions.

Several investigational agents are under evaluation in systemic lupus erythematosus (SLE) clinical trials but quantitative frameworks to enable comparison of their efficacy to reference benchmark treatments are lacking. To benchmark SLE treatment effects and identify clinically important covariates, we developed a model-based meta-analysis (MBMA) within a latent variable model framework for efficacy end points and SLE composite end point scores (BILAG-based Composite Lupus Assessment and Systemic Lupus Erythematosus Responder Index) using aggregate-level data on approved and investigational therapeutics. SLE trials were searched using PubMed and www.

Background: Systolic blood pressure (SBP) is a potential indicator that could guide when to use a resuscitative endovascular balloon occlusion of the aorta (REBOA) in trauma patients with life-threatening injuries. This study aims to determine the optimal SBP threshold for REBOA placement by analyzing the association between SBP pre-REBOA and 24-hour mortality in severely injured hemodynamically unstable trauma patients.

Methods: We performed a pooled analysis of the aortic balloon occlusion (ABO) trauma and AORTA registries.

Systemic lupus erythematosus (SLE) is an autoimmune disease affecting multiple organ systems. Many investigational agents have failed or shown only modest effects when added to standard of care (SoC) therapy in placebo-controlled trials, and only two therapies have been approved for SLE in the last 60 years. Clinical trial outcomes have shown discordance in drug effects between clinical endpoints.

Background: Despite impressive progression-free survival (PFS) results from PARP inhibitors (PARPi) in ovarian cancer, concerns about their effect on post-progression treatment outcomes have recently arisen, particularly when administered in the relapsed setting. Overlapping mechanisms of resistance between PARPi and platinum have been described, and optimal therapies upon progression to PARPi are unknown. We communicate real-world data (RWD) on outcomes of subsequent chemotherapy upon progression to PARPi used as maintenance in ovarian cancer relapses, particularly focusing on platinum rechallenge, according to BRCA status.

Background And Purpose: Perivascular spaces surround the blood vessels of the brain and are involved in neuroimmune functions and clearance of metabolites via the glymphatic system of the brain. Enlarged perivascular spaces could be a marker of dysfunction in these processes and, therefore, are highly relevant to monitoring disease activity in MS. This study aimed to compare the number of enlarged perivascular spaces in people with relapsing MS with MR imaging markers of inflammation and brain atrophy.

Activating RAS missense mutations are among the most prevalent genomic alterations observed in human cancers and drive oncogenesis in the three most lethal tumor types. Emerging evidence suggests mutant KRAS (mKRAS) may be targeted immunologically, but mKRAS epitopes remain poorly defined. Here we employ a multi-omics approach to characterize HLA class I-restricted mKRAS epitopes.

Clinical guidelines promote the identification of several targetable biomarkers to drive treatment decisions in advanced non-small cell lung cancer (NSCLC), but half of all patients do not have a viable biopsy. Specimens from endobronchial-ultrasound transbronchial needle aspiration (EBUS-TBNA) are an alternative source of material for the initial diagnosis of NSCLC, however their usefulness for a complete molecular characterization remains controversial. EBUS-TBNA samples were prospectively tested for several biomarkers by next-generation sequencing (NGS), nCounter, and immunohistochemistry (PD-L1).

Background: Genetic testing for breast cancer susceptibility is widely used, but for many genes, evidence of an association with breast cancer is weak, underlying risk estimates are imprecise, and reliable subtype-specific risk estimates are lacking.

Methods: We used a panel of 34 putative susceptibility genes to perform sequencing on samples from 60,466 women with breast cancer and 53,461 controls. In separate analyses for protein-truncating variants and rare missense variants in these genes, we estimated odds ratios for breast cancer overall and tumor subtypes.

Rationale: Cerebrovascular function is critical for brain health, and endogenous vascular protective pathways may provide therapeutic targets for neurological disorders. S1P (Sphingosine 1-phosphate) signaling coordinates vascular functions in other organs, and S1P (S1P receptor-1) modulators including fingolimod show promise for the treatment of ischemic and hemorrhagic stroke. However, S1P also coordinates lymphocyte trafficking, and lymphocytes are currently viewed as the principal therapeutic target for S1P modulation in stroke.
